Summary: The human colon is home to a diverse microbiome that has been linked to colorectal carcinogenesis. Specific microbial species, such as Fusobacterium nucleatum and Escherichia coli, have been associated with the development of adenoma and adenocarcinoma in the colon. These microorganisms may promote colon carcinogenesis through mechanisms such as adhering to colon cells, modulating gene expression, and interacting with meat components. Moreover, microbiota analysis may have the potential to improve colorectal cancer detection methods and highlight the influence of diet on CRC progression.

Advances are also being made to incorporate microbiome-mediated drug metabolism into drug stability and permeation assays.

Mimicking the dynamic Colonic microbiota in vitro to gain a better understanding on the in vivo metabolism of xenobiotics: Degradation of sulfasalazine

Summary: This study compared the metabolization of the model drug sulfasalazine using the novel dynamic bioreactor MimiCol and a standard static batch fermenter, showing that MimiCol is able to simulate the dynamic conditions found within the ascending colon and lead to an almost 2-fold increase of the metabolization rate constant compared to the static batch fermenter. The study demonstrates that the novel dynamic bioreactor MimiCol can closely simulate physiologically relevant conditions.

This study evaluated the colon drug targeting accuracy of a metronidazole benzoate colonic drug delivery system in healthy human subjects, showing high colon-targeting accuracy and reduced systemic exposure with a non-mucoadhesive formulation.
