Treprostinil palmitil inhibits the hemodynamic and histopathological changes in the pulmonary vasculature and heart in an animal model of pulmonary arterial hypertension

Michel R. Corboz et al.

Summary: Treprostinil palmitil (TP), a long-acting inhaled pulmonary vasodilator prodrug, improves hemodynamics, pulmonary vascular remodeling, and cardiac performance and histopathology in a rat model of pulmonary arterial hypertension (PAH). Its effects are superior to oral sildenafil, suggesting the potential of inhaled TPIS as a treatment for PAH.

Development and Characterization of Treprostinil Palmitil Inhalation Aerosol for the Investigational Treatment of Pulmonary Arterial Hypertension

Adam J. Plaunt et al.

Summary: Researchers have successfully developed a convenient solution-based metered-dose inhaler formulation of TP, known as treprostinil palmitil inhalation aerosol, which demonstrates prolonged efficacy in a rodent model. The new formulation matches the PK profile and efficacy of the traditional TP nebulized formulation, providing a new option for future treatments.

Fabrication of Zein-Lecithin-EGCG complex nanoparticles: Characterization, controlled release in simulated gastrointestinal digestion

Hujun Xie et al.

Summary: Complex nanoparticles of Zein-Lecithin-EGCG were successfully fabricated, showing high encapsulation efficiency and antioxidative capacity. The release of EGCG in simulated gastric and intestinal fluids behaved closely to Fick and First release mechanisms, respectively. The encapsulation of EGCG by Z-L nanocomplexes was achieved through electrostatic, hydrophobic, and hydrogen bonding interactions, leading to excellent stability and sustained release performance for potential applications in the food industry.

Modeling of a spray drying method to produce ciprofloxacin nanocrystals inside the liposomes utilizing a response surface methodology: Box-Behnken experimental design

Isra Khatib et al.

Summary: The study optimized CNL powder production by evaluating the response surface via design of experiments (DoE), finding that sucrose and lactose were superior protectants compared to trehalose. The research demonstrates that the model developed was effective, and the optimum processing conditions for high drug encapsulation involved 57% sucrose, an 80°C inlet temperature, and an atomization rate of 742 L/hr.

Development and Preclinical Evaluation of New Inhaled Lipoglycopeptides for the Treatment of Persistent Pulmonary Methicillin-Resistant Staphylococcus aureus Infections

Adam J. Plaunt et al.

Summary: A customized lipoglycopeptide therapy was developed for chronic pulmonary MRSA disease in cystic fibrosis patients, showing potent activity against MRSA strains in biofilms and within cells. These next-generation compounds exhibit superior pharmacokinetics and are 30 to 60 times more potent than vancomycin in vitro, with RV94 demonstrating the most favorable lung residence time after inhalation in rats. In vivo efficacy of inhaled nebulized RV94 was superior to inhaled vancomycin in a 48-hour acute model of pulmonary MRSA infection in rats.

Intracellular Accumulation of Novel and Clinically Used TB Drugs Potentiates Intracellular Synergy

Lloyd Tanner et al.

Summary: This study focuses on the development of novel therapeutic compounds for drug-resistant tuberculosis, with the phenoxazine PhX1 shown to penetrate murine compartments more effectively than clinically used anti-TB agents and exhibit potential for enhanced drug efficacy. The study also identifies synergies between PhX1 and other compounds, emphasizing the importance of intracellular drug penetration for efficacy. Overall, the data support the utility of the applied assays in identifying in vitro active compounds with potential for clinical development.

Robustness of aerosol delivery of amikacin liposome inhalation suspension using the eFlow® Technology

Zhili Li et al.

Summary: The purpose of the studies was to understand the impact of batch-to-batch variability in the amikacin liposome inhalation suspension (ALIS) formulation and the Lamira nebulizer system on product performance. Results showed that variations in lipid concentration and device hole geometry influenced nebulization time, but critical aerosol performance parameters were maintained within acceptable limits.

Characterisation of cough evoked by inhaled treprostinil and treprostinil palmitil

Richard W. Chapman et al.

Summary: This study investigated the mechanisms of cough induced by TRE and TP, finding that inhaled TRE likely triggers cough by activation of prostanoid IP receptors on jugular C-fibres in the tracheobronchial airways. In rats, TRE administration did not cause apnoea or swallowing, unlike citric acid, capsaicin, and hypertonic saline. Additionally, cough induced by inhaled dry powder and nebulised formulations of TP occurs at higher doses due to the slow release of TRE from the prodrug.
