Neutrophils in cancer: heterogeneous and multifaceted

Catherine C. Hedrick et al.

Summary: Neutrophils have emerged as important regulators of cancer, revealing their diverse and plastic contributions in tumor development and growth, influencing positively and negatively depending on time, place, and disease context. Their functional importance in cancer has been overlooked due to their short-lived, terminally differentiated, and non-proliferative characteristics, but recent studies have led to a paradigm shift in understanding their roles.

Circulating Markers of Neutrophil Extracellular Traps Are of Prognostic Value in Patients With COVID-19

Henry Ng et al.

Summary: Elevated levels of NET markers in patients with moderate to severe COVID-19 were found to be associated with disease severity, respiratory support requirement, and short-term mortality. These markers declined to healthy individual levels four months post-infection. The study suggests a potential role of NETs in COVID-19 disease progression and as targets for novel therapeutic interventions.

Targeting neutrophil extracellular traps with thrombomodulin prevents pancreatic cancer metastasis

Hiroki Kajioka et al.

Summary: Surgery is the primary curative treatment for pancreatic cancer, but postsurgical recurrence is common. Inflammation caused by surgery, neutrophils, and neutrophil extracellular traps (NETs) play important roles in cancer metastasis. Targeting NETs with thrombomodulin may be a novel strategy to improve surgical outcomes for pancreatic cancer patients.

Stromal-driven and Amyloid β-dependent induction of neutrophil extracellular traps modulates tumor growth

Hafsa Munir et al.

Summary: The study reveals that cancer-associated fibroblasts (CAF) induce the formation of neutrophil extracellular traps (NET) within tumors and systemically in the blood and bone marrow. These tumor-induced NETs are driven by a ROS-mediated pathway dependent on CAF-derived Amyloid beta peptide. Inhibition of NETosis skews neutrophils to an anti-tumor phenotype, while t-NETs enhance CAF activation.

TIMP1 Triggers Neutrophil Extracellular Trap Formation in Pancreatic Cancer

Benjamin Schoeps et al.

Summary: The study found that TIMP1 is associated with neutrophil activation in PDAC patients and directly triggers the formation of NETs, impacting the progression of PDAC. By combining plasma levels of TIMP1 and NETs, different prognostic subgroups of PDAC patients can be better identified.

Neutrophil extracellular traps promote gastric cancer metastasis by inducing epithelial-mesenchymal transition

Tong Zhu et al.

Summary: Neutrophil extracellular traps (NETs) in gastric cancer (GC) are associated with tumor progression, metastasis risk, and tumor microenvironment. The study found that NETs formation is affected by the tumor microenvironment and may exacerbate GC progression through promoting epithelial-mesenchymal transition (EMT). Hence, targeting NETs may be therapeutically beneficial in GC treatment.

Role of neutrophil extracellular traps in radiation resistance of invasive bladder cancer

Surashri Shinde-Jadhav et al.

Summary: Radioresistance in bladder cancer treatment is linked to the recruitment of neutrophil extracellular traps (NETs) to the tumor immune microenvironment post-radiation therapy, as demonstrated in a mouse model. Inhibition of NETs can improve radiation response, suggesting NETs as a potential therapeutic target to enhance radiation efficacy.

The Crosstalk Between Cancer Cells and Neutrophils Enhances Hepatocellular Carcinoma Metastasis via Neutrophil Extracellular Traps-Associated Cathepsin G Component: A Potential Therapeutic Target

Xiangqian Guan et al.

Summary: This study revealed that the release of NETs is increased in HCC patients, especially those with portal vein tumor thrombosis. The presence of NETs in HCC tumor tissues is closely associated with a poor prognosis. Functionally, co-culture of HCC neutrophils enhances the invasion ability of HCC cells through NETs formation, while neutrophils from healthy donors inhibit invasion ability. Mechanistic investigations showed that HCC cells-derived cytokine IL-8 triggers NADPH oxidase-dependent NETs formation, promoting HCC metastasis in vitro and in vivo.

Summary: The study shows an increased number of Tregs in NASH-HCC, and depleting Tregs can inhibit the initiation and progression of HCC. There is a positive correlation between increased NETs and hepatic Tregs levels, and NETs promote Treg activity through metabolic reprogramming, linking innate and adaptive immunity. Therapies targeting NETs and Treg interactions may offer a potential strategy for preventing HCC in NASH patients.

DDR1-induced neutrophil extracellular traps drive pancreatic cancer metastasis

Jenying Deng et al.

Summary: The study reveals that DDR1 activation in cancer cells induced by collagen is a major stimulus for the production of CXCL5, leading to the recruitment of TANs and formation of NETs, promoting cancer cell invasion and metastasis in PDAC. Furthermore, the collagen-induced CXCL5 production is mediated by a DDR1/PKC theta/SYK/NF-kappa B signaling cascade.
