Discordance of immunotherapy response predictive biomarkers between primary lesions and paired metastases in tumours: A systematic review and meta-analysis

Yutian Zou et al.

Summary: This study included 56 studies and found a high discordance rate for PD-L1, PD-1, PD-L2, and TIL levels, while TMB and MSI status were less likely to change between primary tumors and paired metastases.

Immuno-Oncological Biomarkers for Squamous Cell Cancer of the Head and Neck: Current State of the Art and Future Perspectives

Stijn J. De Keukeleire et al.

Summary: Squamous cell cancer of the head and neck (SCCHN) is a common malignancy with poor prognosis, and research has focused on diagnostic, prognostic, and predictive biomarkers. Despite the importance of PD-L1 expression as a predictor, there are still technical challenges and standardization issues to be addressed. Novel biomarkers and exploration of the tumor microenvironment continue to provide insights for diagnosis and treatment of SCCHN.

Oropharyngeal Squamous Cell Carcinoma Treatment in the Era of Immune Checkpoint Inhibitors

Peter L. Stern et al.

Summary: While head and neck squamous cell carcinomas (HNSCC) are decreasing due to reduced exposure to tobacco and alcohol, high-risk HPV-positive OPSCC, especially in the tonsil and base of tongue subsites, are increasing. Understanding the integration of immune factors in the TME may lead to identifying prognostic biomarkers and novel therapeutic strategies. The use of immune checkpoint inhibitors, specifically the PD-1/PD-L1 pathway, for HPV-positive OPSCC treatment shows potential for being less toxic and more effective.

Recurrent HNSCC Harbor an Immunosuppressive Tumor Immune Microenvironment Suggesting Successful Tumor Immune Evasion

Summary: Recurrent tumors of head and neck squamous cell carcinoma have an immunosuppressive tumor immune microenvironment, particularly apparent after adjuvant chemoradiotherapy, which may substantially impact therapeutic response and prognosis.
