Ex vivo culture of intact human patient derived pancreatic tumour tissue

John Kokkinos et al.

Summary: This study developed a novel ex vivo explant model that maintains the 3D architecture and microenvironmental cues of human pancreatic ductal adenocarcinoma (PDAC). The model showed advantages over standard organoids, retaining functional multi-cellular stroma and fibrosis, providing unprecedented opportunities for studying PDAC biology and personalized treatment.

Progress towards non-small-cell lung cancer models that represent clinical evolutionary trajectories

Robert E. Hynds et al.

Summary: NSCLC is the leading cause of cancer-related deaths worldwide, and although progress is being made in early detection and the development of targeted therapies and immunotherapies, the survival rate for advanced patients remains low. Effective cancer research relies on pre-clinical models that accurately reflect disease progression and patient responses to therapy.

Development of a patient-derived explant model for prediction of drug responses in endometrial cancer

Anna Collins et al.

Summary: This study developed a novel PDE model system for endometrial cancer to monitor drug responses in a pre-clinical setting. Results showed that EC samples are amenable to PDE culture with preserved histological architecture and viability for up to 48 hours. The PDE platform demonstrated differential responses to conventional chemotherapeutics and immune checkpoint inhibition, providing evidence for personalized drug-testing in a pre-clinical setting.

Human tissue cultures of lung cancer predict patient susceptibility to immune-checkpoint inhibition

David Junk et al.

Summary: This study introduces a standardized ex vivo tissue culture model to investigate individual responses to immunotherapies in non-small cell lung cancer (NSCLC) patients. The model demonstrates the functionality of T cells in NSCLC slice cultures up to 10 days ex vivo, suggesting its potential for patient stratification and in-depth investigation of tumor-associated T-cell regulation.

Summary: The study outlines the development of a colorectal cancer ex vivo model using fresh tumor samples, highlighting the preservation of original tumor genetic features and partial retention of architecture and non-malignant cells in culture. The model shows promise for use in pre- and co-clinical settings.

Queensland Genomics: an adaptive approach for integrating genomics into a public healthcare system

Miranda E. Vidgen et al.

Summary: The implementation of genomics in healthcare systems presents challenges which can be addressed by drawing lessons from other programs. An adaptive approach allows for flexibility to respond to learnings and external factors. Challenges include engagement of people and services, and the design of individual projects and the overall program.

Toward improved in vitro models of human cancer

Jose M. Ayuso et al.

Summary: Cancer remains a leading cause of death worldwide with limited treatment options for certain tumors. The inefficiencies of traditional in vitro tools and animal models in predicting drug sensitivity contribute to the challenges in drug development. Patient-specific BMOMs show promise in improving our understanding of cancer and predicting patient response to therapy.

Patient-derived tumour models for personalized therapeutics in urological cancers

Arjanneke F. van de Merbel et al.

Summary: Preclinical knowledge and potential biomarkers for urological cancers have limited translation into clinical practice. Patient-derived tumor models, including 3D culture systems and xenograft models, have improved capacity to mimic the complexity of urological cancers. Opportunities for personalized therapeutic approaches for patients with urological cancers are limited and clinical validation of preclinical data is crucial for translation into treatment decisions.
