Whole-body magnetic resonance imaging for prostate cancer assessment: Current status and future directions

Sandy Van Nieuwenhove et al.

Summary: Over the past decade, advancements in updated definitions and therapies have remarkably changed the management of prostate cancer patients. Modern imaging techniques, including whole-body magnetic resonance imaging (WB-MRI) and nuclear medicine, provide accurate staging and evaluation of disease response to treatment. This paper discusses the evolution and current state-of-the-art of WB-MRI imaging protocols and highlights its applications in prostate cancer care.

aPROMISE: A Novel Automated PROMISE Platform to Standardize Evaluation of Tumor Burden in 18F-DCFPyL Images of Veterans with Prostate Cancer

Nicholas Nickols et al.

Summary: Standardized staging and quantitative reporting are necessary in F-18-DCFPyL PET/CT imaging for demonstrating clinical outcomes. This study introduces an automated platform called aPROMISE to implement and extend the PROMISE criteria for evaluating prostate cancer. The performance of aPROMISE in staging and quantifying disease burden was validated in prostate cancer patients who underwent PSMA imaging. The results show that aPROMISE is consistent between readers and can lead to upstaging in PSMA imaging.

Summary: PSMA PET/CT can be used to evaluate treatment response in patients with metastatic prostate cancer before and after any local and systemic treatment. Ideally, imaging criteria should categorize patients as responders, patients with stable disease, partial response, and complete response, or as non-responders. Specific clinical scenarios deserve special consideration.

Summary: This study compared the prognostic value of an artificial intelligence-based method for estimating Total Metabolic Tumor Volume (TMTV) with TMTV measured by experts in diffuse large B-cell lymphoma (DLBCL) patients. Incorporating radiomics into clinical trials: expert consensus on considerations for data-driven compared to biologically driven quantitative biomarkers

Summary: Existing quantitative imaging biomarkers go through a well-understood validation process before being included in clinical trials, while radiomics extracts features from imaging data without prior assumptions. The selection and incorporation of relevant features in radiomics require additional considerations for meaningful imaging endpoints. The large number of radiomic features tested may lead to sample sizes impractical within clinical trials.

Summary: This study establishes a novel methodology using experimental ultrasound techniques to assess the morphological, functional, and molecular features of breast cancer bone metastases, compared with magnetic resonance imaging and histological analysis. The results show that ultrasound techniques can effectively characterize osteolytic lesions and that molecular ultrasound allows for the non-invasive assessment of increased VEGFR2 expression in skeletal lesions.

Impact of 18F-FDG PET, PET/CT, and PET/MRI on Staging and Management as an Initial Staging Modality in Breast Cancer A Systematic Review and Meta-analysis

Sangwon Han et al.

Summary: The systematic review and meta-analysis found that the use of F-18-FDG PET, PET/CT, or PET/MRI during the initial staging of breast cancer can significantly modify staging and treatment plans for patients. Factors such as age, initial stage, and histologic grade were identified as significant factors affecting the heterogeneity in changes in stage or management. Routine clinical use of PET imaging for initial staging of breast cancer may be beneficial based on current literature.

[18F]FDG PET/CT quantitative parameters for the prediction of histological response to induction chemotherapy and clinical outcome in patients with localised bone and soft-tissue Ewing sarcoma

Alessio Annovazzi et al.

Summary: This study aimed to evaluate the diagnostic accuracy of [F-18]FDG PET/CT volumetric parameters in predicting response to chemotherapy and clinical outcome in patients with EWS. The results showed that Delta TLG was the best predictor for histologic response, while SUV2 and Delta TLG were independent predictors of worse progression-free survival. The study suggests that [F-18]FDG PET/CT could accurately predict histologic response to neoadjuvant chemotherapy and may have prognostic value for future disease relapse in patients with EWS.
