Type I interferon activates MHC class I-dressed CD11b+ conventional dendritic cells to promote protective anti-tumor CD8+ T cell immunity

Ellen Duong et al.

Summary: In this study, the researchers investigated the different roles of tumor-infiltrating dendritic cells (DCs) in anti-tumor immunity. By comparing spontaneously regressing and progressing tumors, they found that tumor-reactive CD8+ T cells were lost in progressing tumors but preserved in regressing tumors. They also identified a specific type of DCs (ISG(+) DCs) in regressing tumors that activated CD8+ T cells and played an important role in presenting tumor-derived antigens. Furthermore, they showed that exogenous IFN-b stimulation could restore the lost anti-tumor immunity in progressing tumors. These findings suggest that targeting this functional DC state may be a potential approach for the treatment of human diseases.

Tumor-infiltrating plasmacytoid dendritic cells are associated with survival in human colon cancer

Maximilian Kiessler et al.

Summary: The study found that higher densities of tumor-infiltrating pDCs are associated with lower UICC staging, increased progression-free and overall survival in colon cancer patients. A lower number of colon cancer-infiltrating pDCs was independently linked to worse prognosis, while the colocalization of activated pDCs and T cells in tumor stroma and within TLS may contribute to the correlation between higher pDC densities and better prognosis.

Summary: Current cancer immunotherapy efforts focus on enhancing T cell effector function, but stimulating dendritic cells (DCs) may improve treatment outcomes. Specifically, utilizing the Flt3/Flt3L axis to boost DC function shows promising potential for cancer immunotherapy.

DC-Derived Exosomes for Cancer Immunotherapy

Yi Yao et al.

Summary: Dendritic cells (DCs)-based cancer vaccines have not been very successful due to tumor-mediated immunosuppression, leading to limited clinical efficacy. DC-derived exosomes (DCexos) are considered as a promising alternative to DC-based vaccines, as they are more resistant to tumor-mediated suppression and have shown better anti-tumor efficacy in pre-clinical models.

Type 2 dendritic cells mediate control of cytotoxic T cell resistant tumors

Stephen Iwanowycz et al.

Summary: DC2s, the majority of conventional DCs within most tumors, have the potential to initiate and sustain antitumor immunity, showing significant correlation with survival across multiple human cancers. Impairment of DC1s and enhancement of DC2 functionality leads to improved tumor control, highlighting the therapeutic and prognostic potential of DC2s within checkpoint blockade-resistant tumors.
