Mesenchymal stem cells and cancer therapy: insights into targeting the tumour vasculature

Surendar Aravindhan et al.

Summary: The interaction between the tumor microenvironment and tumor cells plays a crucial role in tumor progression, with mesenchymal stem cells (MSCs) being key players in modulating the tumor microenvironment and influencing the fate of tumor cells. MSCs exhibit dual functions in the tumor microenvironment, either promoting tumorigenicity or inhibiting tumor growth. Their ability to release mediators such as exosomes and migrate to tumor sites facilitates efficient drug delivery and targeting of migrating tumor cells.

CAR-NK cells from engineered pluripotent stem cells: Off-the-shelf therapeutics for all patients

Shi-Jiang Lu et al.

Summary: The clinical success of CAR-T therapy for hematological malignancies has led to significant advancements in cellular immunotherapy. However, challenges such as affordability, availability, and scientific optimization still exist. CAR-NK cells, derived from NK cells which do not require HLA restriction, offer a promising off-the-shelf therapeutic option for patients. PSCs can be engineered with CARs to produce unlimited and homogeneous CAR-NK cells for various disease targets. Additionally, a 3D bioreactor platform for PSC expansion and NK cell production addresses key barriers related to cost and scalability for CAR-NK therapy.

Natural Killer Cells Generated From Human Induced Pluripotent Stem Cells Mature to CD56brightCD16+NKp80+/- In-Vitro and Express KIR2DL2/DL3 and KIR3DL1

Johanna Euchner et al.

Summary: The study focused on generating CD56(+)CD16(+)CD3(-) NK lymphocytes from hiPSC, and characterized the development of NK cells by examining the surface expression of NK-lineage markers. Through hematopoietic priming, high purity CD56(bright)CD16(-) and CD56(bright)CD16(+) NK cells were efficiently differentiated. The generated NK cells exhibited functional cytotoxicity, degranulation, and antibody-dependent cell-mediated cytotoxicity, confirming their maturity up to stage 4b.

Generation of hypoimmunogenic T cells from genetically engineered allogeneic human induced pluripotent stem cells

Bo Wang et al.

Summary: The study demonstrates the generation of hypoimmunogenic cancer-antigen-specific T cells derived from human induced pluripotent stem cells, which escape rejection in allogeneic mice due to lacking certain components of the human leukocyte antigen system and incorporating a ligand that inhibits natural killer cells. These T cells also showed resistance to recognition by immune cells and maintained anti-tumor potency, highlighting their potential contribution to off-the-shelf T cell immunotherapies.

DL4-μbeads induce T cell lineage differentiation from stem cells in a stromal cell-free system

Ashton C. Trotman-Grant et al.

Summary: A stromal cell-free, microbead-based approach has been developed to efficiently generate T-lineage cells from various cell sources, including cord blood, GCSF-mobilized peripheral blood, and pluripotent stem cells. The method involves the use of DL4-mu beads and lymphopoietic cytokines to induce a sequential differentiation process leading to the development of functional T cells. Single-cell RNA sequencing of human pluripotent stem cell-derived proT cells shows a transcriptional profile similar to early thymocytes. Adoptive transfer of CD34(+)CD7(+) proT cells into immunodeficient mice demonstrates successful thymic engraftment and maturation of peripheral T cells. The DL4-mu bead platform provides a simple and robust tool for studying human T cell development and developing engineered T cell therapies from sustainable sources.
