Metabolic resistance to the inhibition of mitochondrial transcription revealed by CRISPR-Cas9 screen

Mara Mennuni et al.

Summary: Resistance to mitochondrial transcription inhibitors (IMTs) is mainly achieved through compensatory increase in mitochondrial DNA expression and cellular metabolites. Loss of genes related to von Hippel-Lindau and mTORC1 pathways confers resistance to the inhibitors. Downregulation of mitochondrial transcription factor A and inhibition of mitochondrial translation also impairs survival of resistant cells.

LONP1 and ClpP cooperatively regulate mitochondrial proteostasis for cancer cell survival

Yu Geon Lee et al.

Summary: Mitochondrial proteases LONP1 and ClpP work together to alleviate proteotoxic stress and protect mitochondrial functions, supporting cancer cell survival. They target crucial components of mitochondrial functions and have overlapping substrates, such as SHMT2, to regulate cancer cell growth and sensitivity to metabolic stress. Patients with higher expression levels of LONP1 and ClpP have poorer prognosis in prostate cancer, suggesting that targeting the mitochondrial proteostasis network via these proteases could be a potential therapeutic strategy.

Receptor-Mediated Mitophagy Rescues Cancer Cells under Hypoxic Conditions

Alibek Abdrakhmanov et al.

Summary: Stimulation of mitochondria quality control by hypoxia-induced mitophagy, involving BNIP3 and BNIP3L proteins, can contribute to tumor cell resistance to apoptosis stimuli. Targeting mitochondria with TTFA in normoxia enhances cisplatin-induced apoptosis, while in hypoxia, cells survive through receptor-mediated mitophagy, accumulating BNIP3 and BNIP3L. This suggests that receptor-mediated mitophagy plays a crucial role in cell death suppression under hypoxic conditions.

Mitochondrial Protein Translation: Emerging Roles and Clinical Significance in Disease

Fei Wang et al.

Summary: Mitochondria in cells are crucial organelles with their own genetic system, and their protein translation process is regulated by various factors that, when abnormal, can lead to diseases. The balance between mitochondrial protein translation and cytoplasmic translation is also important for physiological and pathological processes in the body.
