Use of artificial intelligence to enhance phenotypic drug discovery

Sophia Malandraki-Miller et al.

Summary: Research and development productivity in the pharmaceutical industry has been closely scrutinised, with phenotypic and target-based approaches holding different advantages but facing challenges in clinical translation. The adoption of artificial intelligence and machine learning tools shows promise for revolutionising drug development, while public-private partnerships and interdisciplinary collaborations are key for fostering innovation and efficient drug discovery.

Article Biochemistry & Molecular Biology

Deficient LEF1 expression is associated with lithium resistance and hyperexcitability in neurons derived from bipolar disorder patients

Renata Santos et al.

Summary: The study revealed that the activity of the Wnt/β-catenin signaling pathway was severely affected and the expression of LEF1 was significantly decreased in Li-non-responsive neurons, potentially contributing to lithium resistance. Valproic acid upregulated LEF1, reducing excitability in Li-non-responsive neurons.

Summary: A comprehensive and complex automated platform for high throughput screenings (HTS) in a translational research setting is described, which is designed for maintenance and expansion of different cell types. The platform allows automation of all cultivation steps and is flexible for development of methods for variable cell types.

Altered network and rescue of human neurons derived from individuals with early-onset genetic epilepsy

Priscilla D. Negraes et al.

Summary: The study utilized induced pluripotent stem cells derived from individuals deficient in CDKL5 protein to generate neural cells, revealing proliferation defects in neural progenitor cells, morphological alterations and compromised glutamatergic synaptogenesis in neurons. Furthermore, electrical activity of CDD cortical neurons showed hyperexcitability during development, leading to an overly synchronized network.

Drug Repurposing for Rare Diseases

Helen Roessler et al.

Summary: There are around 7000 identified rare diseases, with less than 6% having an approved treatment option. Drug repurposing is a time-saving and cost-efficient method that can significantly reduce the risk of drug development for rare diseases. New strategies have been developed in recent years for a systematic and rational approach to drug repurposing.
