Molecular regulation of neuroinflammation in glaucoma: Current knowledge and the ongoing search for new treatment targets

Gulgun Tezel

Summary: Neuroinflammation plays a significant role in the neurodegenerative processes of glaucoma, with glial cells and peripheral immune cells causing toxicity to retinal ganglion cells. The interaction between glial cells and immune cells is crucial in amplifying neuroinflammation and determining the neurodegenerative outcome. Inhibition of neurodegenerative inflammation may provide a broad treatment strategy for rescuing retinal ganglion cell integrity and function, while targeting both inflammatory toxicity and mitochondrial dysfunction.

Retina in a dish: Cell cultures, retinal explants and animal models for common diseases of the retina

Sven Schnichels et al.

Summary: The exact pathogenesis of retinal diseases like AMD, glaucoma, and DR remains unclear, and current therapeutic options are often unsatisfactory. Research heavily relies on animal models, but there is pressure to reduce animal research. Improved disease models are needed to understand pathologies and develop treatment options that require fewer or no experimental animals.

Increased ratios of complement factors C3a to C3 in aqueous humor and serum mark glaucoma progression

W. H. G. Hubens et al.

Summary: The study found that there were significant increases in the AH and serum complement C3a/C3 ratios in progressive POAG patients, which correlated strongly with the rate of disease progression. This suggests that complement system activation may play a role in glaucoma progression.

Activation of Apoptosis in a βB1-CTGF Transgenic Mouse Model

Maximilian Weiss et al.

Summary: In this study, utilizing the βB1-CTGF mouse model to investigate the pathomechanisms of glaucoma revealed that apoptotic processes and synapse loss precede neuronal death in this model. The slow progression rate of degeneration in the βB1-CTGF mice makes them a suitable model for studying primary open-angle glaucoma.

Age-related macular degeneration

Monika Fleckenstein et al.

Summary: Age-related macular degeneration (AMD) is the leading cause of legal blindness in the industrialized world, with a complex pathophysiology involving aging, environmental factors, and genetic susceptibility leading to photoreceptor degeneration and drusen formation. Tailoring therapeutics to specific phenotypes and stages may be crucial for preventing irreversible vision loss in the future.

Reduced Retinal Degeneration in an Oxidative Stress Organ Culture Model through an iNOS-Inhibitor

Ana M. Mueller-Buehl et al.

Summary: The study investigated the potential of an inducible nitric oxide synthase (iNOS) inhibitor to prevent oxidative stress in retinal organ cultures, which is relevant to the development of various retinal diseases including glaucoma. Results showed that the inhibitor was able to prevent degeneration in porcine retinas, offering a promising treatment option for retinal diseases.
