Cofilin-1, LIMK1 and SSH1 are differentially expressed in locally advanced colorectal cancer and according to consensus molecular subtypes

Annie Cristhine Moraes Sousa-Squiavinato et al.

Summary: CFL-1 and its modulators, LIMK1/SSH1, exhibit differential expression and are associated with lymph node metastasis in colorectal cancer (CRC). Upregulation of LIMK1 is linked to lower overall survival rates and greater lymph node metastatic potential in all tumor stages, while downregulated expression of CFL-1 and SSH1 is associated with greater lymph node metastatic potential and deeper local invasion. Furthermore, LIMK1 and SSH1 genes are upregulated in immune (CMS1) and mesenchymal (CMS4) subtypes of CRC, with high levels of LIMK1 also correlating with lower overall survival rates in canonical (CMS2) and metabolic (CMS3) subtypes.

Conformational plasticity of the ULK3 kinase domain

Sebastian Mathea et al.

Summary: The human protein kinase ULK3 plays a role in regulating membrane abscission, exosome budding, and cytokinesis. The high resolution structures of ULK3 kinase domain reveal its unique features and potential as a druggable target. The study also demonstrates a conformational switch between active and inactive states of ULK3, controlled by small-molecule binders, and proposes a potential substrate-recognition mechanism for full-length ULK3 protein.
