DIVERSITY AND BIOLOGY OF CANCER-ASSOCIATED FIBROBLASTS

Giulia Biffi et al.

Summary: Efforts to develop anti-cancer therapies have mainly targeted the epithelial compartment, but recent studies have shown the significant influence of cancer-associated fibroblasts (CAFs) in tumor progression. CAFs not only promote cancer cell proliferation, therapy resistance, and immune exclusion, but may also restrain tumor progression in certain contexts. Research on CAFs has focused on their heterogeneity, plasticity, and functions across different cancer types and states, as well as advancements in therapeutic strategies targeting CAFs currently undergoing preclinical and clinical evaluation.

Leukemia inhibitory factor is a novel biomarker to predict lymph node and distant metastasis in pancreatic cancer

Wenna Jiang et al.

Summary: LIF overexpression in pancreatic tumor tissues is associated with poorer overall survival and recurrence-free survival. Higher serum LIF levels in pancreatic cancer patients are more effective in predicting lymph node and distant metastasis compared to other biomarkers.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

Inhibition of Hedgehog Signaling Alters Fibroblast Composition in Pancreatic Cancer

Nina G. Steele et al.

Summary: The study reveals that Hedgehog signaling is uniquely activated in fibroblasts in PDAC, with varying levels of elevation in myofibroblastic CAFs compared to inflammatory CAFs. Inhibition of the Hedgehog pathway slows tumor growth, alters the composition of CAF subtypes, and impacts immune cell infiltration, leading to increased immunosuppression.

Katherine S. Yang et al.

Summary: By analyzing extracellular vesicles (EV) in peripheral blood from patients with intraductal papillary mucinous neoplasms (IPMNs), it was found that MUC5AC can predict the presence of invasive carcinoma within IPMN. This approach has the potential to improve management and follow-up of IPMN patients, including avoiding unnecessary surgeries.

Type I collagen deletion in αSMA+ myofibroblasts augments immune suppression and accelerates progression of pancreatic cancer

Yang Chen et al.

Summary: The stromal desmoplastic reaction in pancreatic ductal adenocarcinoma involves significant accumulation of type I collagen (Col1). Deletion of Col1 specifically in myofibroblasts accelerates PDAC progression by increasing Cxcl5 expression in cancer cells and promoting recruitment of myeloid-derived suppressor cells. Targeting CXCR2 and CCR2 may help restrain accelerated PDAC progression in the absence of stromal Col1.

Mutant KRAS Downregulates the Receptor for Leukemia Inhibitory Factor (LIF) to Enhance a Signature of Glycolysis in Pancreatic Cancer and Lung Cancer

Suhu Liu et al.

Summary: Pancreatic cancer is often driven by the oncogenic KRAS mutation, which leads to downregulation of the LIF receptor (LIFR) and subsequent increased expression of GLUT1, promoting enhanced glycolysis and cellular transformation. The LIFR/STAT3 pathway may have tumor-suppressive effects on cancer cells with mutated KRAS, indicating a potential therapeutic target for treating KRAS-driven malignancies.

An engineered ligand trap inhibits leukemia inhibitory factor as pancreatic cancer treatment strategy

Sean A. Hunter et al.

Summary: Hunter et al. engineered a high affinity, soluble variant of leukemia inhibitory factor receptor (LIFR) to serve as a ligand trap for the LIF cytokine. They demonstrated that this engineered LIFR exhibits improved affinity relative to the wild-type receptor, leading to better disruption of LIF signaling in cancer cells, and highlighting promise of such ligand traps as therapeutic strategy for cancer treatment.

Zinc Finger E-Box Binding Homeobox 1 Promotes Cholangiocarcinoma Progression Through Tumor Dedifferentiation and Tumor-Stroma Paracrine Signaling

Cindy Lobe et al.

Summary: The transcription factor ZEB1 plays a crucial role in the progression of cholangiocarcinoma by regulating the crosstalk between tumor cells and cancer-associated fibroblasts (CAFs), leading to tumor dedifferentiation and CAF activation. In vitro and in vivo experiments demonstrated that ZEB1 promotes epithelial-mesenchymal transition and stemness in tumor cells, as well as inducing myofibroblast proliferation through the upregulation of connective tissue growth factor (CTGF). The study also found that ZEB1 controls the expression of paracrine signals such as HGF and IL6 in both tumor cells and myofibroblasts.

Epigenetic and Transcriptional Control of the Epidermal Growth Factor Receptor Regulates the Tumor Immune Microenvironment in Pancreatic Cancer

Summary: This study identifies lysine demethylase 3A (KDM3A) as an epigenetic regulator that affects the response to immunotherapy in pancreatic ductal adenocarcinoma by modulating the expression of epidermal growth factor receptor (EGFR) through Krueppel-like factor 5 (KLF5) and SMAD family member 4 (SMAD4). Inhibition of EGFR can reshape the immune microenvironment, offering a potential immunotherapy-sensitizing strategy for PDA.
