Understanding the organ tropism of metastatic breast cancer through the combination of liquid biopsy tools

Lorenzo Gerratana et al.

Summary: The study explored the combination of circulating tumour DNA (ctDNA) analysis and circulating tumour cells (CTCs) enumeration in estimating target organs more susceptible to metastatic breast cancer (MBC) involvement. Results suggest that ctDNA and CTC enumeration could provide useful insights regarding MBC organotropism, indicating a possible role for future monitoring strategies focusing on high-risk organs defined by tumour biology.

Circulating tumor DNA in neoadjuvant-treated breast cancer reflects response and survival

M. J. M. Magbanua et al.

Summary: The study investigated the utility of serial ctDNA testing in predicting pCR and risk of metastatic recurrence during neoadjuvant chemotherapy for high-risk early breast cancer patients. The results showed that lack of ctDNA clearance was a significant predictor of poor response and metastatic recurrence, while clearance was associated with improved survival even in patients who did not achieve pCR. Personalized monitoring of ctDNA during NAC may aid in real-time assessment of treatment response and help fine-tune pCR as a surrogate endpoint of survival.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Circulating tumor DNA dynamics in advanced breast cancer treated with CDK4/6 inhibition and endocrine therapy

Olga Martinez-Saez et al.

Summary: This study found that ctDNA dynamics using a standardized multi-gene panel and a unique methodological approach can predict treatment outcome in patients with advanced breast cancer. Specifically, calculating the mean of variant allele fraction ratios for detected mutations was significantly associated with progression-free survival.

Circulating Tumor DNA as a Predictive Marker of Recurrence for Patients With Stage II-III Breast Cancer Treated With Neoadjuvant Therapy

Po-Han Lin et al.

Summary: The presence of ctDNA after neoadjuvant therapy is a robust marker for predicting relapse in stage II to III breast cancer patients. Advanced nodal status and ctDNA positivity after NAT were significant risk factors for recurrence.

Summary: The study evaluates early metabolic responses to neoadjuvant trashizumab and pertuzumab using F-18-FDG-PET and the possibility of chemotherapy de-escalation using a pathological response-adapted strategy in patients with HER2-positive, early-stage breast cancer.

Genomic profile of advanced breast cancer in circulating tumour DNA

Belinda Kingston et al.

Summary: Genomic profiling of advanced breast cancer using plasma circulating tumour DNA sequencing revealed diverse subclonal resistance mutations, with distinct mutational processes identified in ER-positive breast cancer. This study highlights the importance of subclonal diversification in pre-treated advanced breast cancer, presenting novel therapeutic opportunities.

Association of 17q22 Amplicon Via Cell-Free DNA With Platinum Chemotherapy Response in Metastatic Triple-Negative Breast Cancer

Katharine A. Collier et al.

Summary: The 17q22 amplicon detected in ctDNA is associated with improved progression-free survival with platinum chemotherapy in patients with metastatic triple-negative breast cancer. However, there was no association between 17q22 amplification and PFS in patients receiving non-platinum chemotherapy agent capecitabine.
