p62/SQSTM1 in liver diseases: the usual suspect with multifarious identities

Chong Teik Tan et al.

Summary: Sequestosome-1 (SQSTM1), also known as p62, is an autophagy receptor that plays a crucial role in the pathogenesis of liver diseases such as NASH and HCC. Abundance of p62 inclusion bodies has been recognized as a biomarker for these diseases. However, the underlying mechanisms by which p62 regulates and contributes to the development and progression of these diseases remain incompletely understood. This review critically summarizes the findings from preclinical models and explores the potential of p62 as a predictive biomarker and therapeutic target for liver diseases.

Systematic Analysis of Cytostatic TGF-Beta Response in Mesenchymal-Like Hepatocellular Carcinoma Cell Lines

Medine Zeynep Gungor et al.

Summary: The mesenchymal-like HCC cell lines showed resistance to TGF-beta-induced growth suppression. Most cell lines had active canonical signaling from the cell membrane to the nucleus, while three cell lines lost expression of cytostatic effector genes. This suggests a selective loss of cytostatic TGF-beta responses in mesenchymal-like HCC cell lines, independent of impairment of the TGF-beta signaling cascade, making them valuable models for studying molecular mechanisms in hepatocarcinogenesis.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.
