Very low mutation burden is a feature of inflamed recurrent glioblastomas responsive to cancer immunotherapy

Matthias Gromeier et al.

Summary: The study found that low tumor mutation burden is associated with longer survival after immunotherapy in recurrent glioblastoma patients, and this relationship is related to inflammatory gene signatures, which evolves upon recurrence.

The Challenges of Tumor Mutational Burden as an Immunotherapy Biomarker

Denis L. Jardim et al.

Summary: Tumor mutational burden (TMB) reflects cancer mutation quantity and correlates with better outcomes of immune checkpoint inhibitors (ICIs). However, TMB as a biomarker is imperfect and a composite predictor including critical variables is needed.

Diffuse Glioma Heterogeneity and Its Therapeutic Implications

James G. Nicholson et al.

Summary: Diffuse gliomas are characterized by high levels of genetic, epigenetic, and environmental heterogeneity, leading to extreme phenotypic heterogeneity at the cellular level and resistance to various therapies. The intratumoral heterogeneity and malignant cell state plasticity in gliomas pose significant challenges to the development of novel targeted therapies.

Genomes for Kids: The Scope of Pathogenic Mutations in Pediatric Cancer Revealed by Comprehensive DNA and RNA Sequencing

Scott Newman et al.

Summary: This study utilized a three-platform sequencing approach, including WGS, WES, and RNAseq, to examine tumor and germline genomes from 309 children with pediatric cancers. The study revealed the presence of diverse genetic variants in most patients, with WGS helping to detect activating gene fusions, enhancer hijacks, small intragenic deletions, and mutational signatures. The evaluation of paired tumor-normal data showed the relevance of pathogenic germline variants to tumor development in a significant portion of cases.
