Whole-exome sequencing in eccrine porocarcinoma indicates promising therapeutic strategies

Summary: The study of the mutational landscape of EP revealed a high overall mutation rate attributed to processes related to UV exposure, APOBEC enzyme dysregulation, and defective homologous double-strand break repair. Recurrent driving somatic alterations were detected in EP candidate driver genes and chromosomal regions, with a reduction or loss of BRCA2 protein in 50% of EP tumors. The results suggest that defective homologous double-strand break repair and the p53 pathway play important roles in EP etiology, indicating potential treatment strategies targeting these pathways.

These carcinomas are aggressive with high chances of local recurrence and distant metastasis, and assessment of Wnt signaling pathway components may be useful for diagnostic and targeted therapy.
