Summary: The majority of in vitro cancer research studies are conducted in 2D and subsequently validated in animal models, but both models have limitations. The proposed solution to address these discrepancies is the use of 3D culture models, which have been shown to exhibit changes in cell behavior compared to 2D models. These changes include proliferation, gene expression, and chemosensitivity, and can provide better comparability to in vivo cells.

3D printing novel in vitro cancer cell culture model systems for lung cancer stem cell study

Summary: Traditional two-dimensional (2D) in vitro cell cultures and animal models are limited in simulating the native environment for cancer stem cell (CSC) studies, but novel three-dimensional (3D) cell cultures show promise in enriching CSCs from lung cancer patients. Different 3D printing platforms were used to produce rigid and hydrogel-based scaffolds, with gene expression analysis confirming overexpression of stemness and invasion promoters in cells grown on hydrogel scaffolds and tumor spheres. The 3D printed models, especially those using GelMA-PEGDA hydrogels, outperform 2D culture models in mimicking tumor complexity and regulating cancer cell behavior.

Vimentin served as a biomarker for predicting brain metastasis and poor prognosis in these patients. The EMT pathway may be a potential therapeutic target in high-risk populations.
