Pseudoexfoliation syndrome and glaucoma: from genes to disease mechanisms

Summary: Recent research has identified seven loci associated with the risk of PEX that surpass genome-wide significance. The main genetic effect locus is LOXL1, with common risk variants showing reversed effects in different ethnicities. Other PEX-associated genetic variants have provided insights into additional disease processes and pathways, including ubiquitin-proteasome function, calcium signaling, and lipid biosynthesis. Integration of multiomics data has further contributed to understanding the complex etiology underlying PEX syndrome and glaucoma.

The results showed that iris volume did not differ significantly in PXF and PXG eyes, but both had significantly greater volume compared to POAG eyes. Iris parameters or TM length were not correlated with clinical variables such as IOP, age, or visual field indices.
