Raj Kamal et al.

Summary: In this study, eleven novel [1,2,4]triazolo[4,3-a]pyrimidines were synthesized efficiently using DIB as the hypervalent iodine(III) reagent, and the regiochemistry of the final products was established through single crystal and X-ray crystallographic analysis. The synthesized compounds showed significant cytotoxicity against human osteosarcoma bone cancer and breast cancer cell lines, with compound 2g identified as a potent apoptotic inducer.

Visible-Light-Induced Formation of Thiavinyl 1,3-Dipoles: A Metal-Free [3+2] Oxidative Cyclization with Alkynes as Easy Access to Thiophenes

Baihui Zheng et al.

Summary: A visible-light-induced [3+2] oxidative cyclization of alkynes with ketene dithioacetals in the presence of an acridine photosensitizer was reported, leading to regioselective synthesis of multisubstituted thiophenes in high yields under metal-free conditions. This reaction showed good substrate tolerance and efficiency in large-scale syntheses. The reaction mechanism and applications were detailed to illustrate the reactivity of the new 1,3-dipoles and the selectivity of the reactions.

Summary: A metal-free approach for the synthesis of seven-membered N-heterocycles has been developed using I-2-promoted intramolecular cross-coupling/annulation of butenyl anilines. This reaction involves C-H activation and C-C bond formation and shows good functional group tolerance, leading to a series of benzo[b]azepine derivatives in moderate to good yields.

Summary: The reaction of flavanones with hypervalent iodine reagents was found to lead to the synthesis of isoflavones and flavones, with the former resulting from an oxidative 2,3-aryl rearrangement. However, the overall synthetic utility and generality of the reaction may be more limited than previous literature reports suggest.

Stereoselective Oxidative Cyclization of N-Allyl Benzamides to Oxaz(ol)ines

Ayham H. Abazid et al.

Summary: This study introduces an enantioselective oxidative cyclization method for N-allyl carboxamides using a chiral triazole-substituted iodoarene catalyst, yielding highly enantioenriched oxazolines and oxazines with up to 94% yield and 98% ee enantioselectivity. The method is versatile, allowing the construction of quaternary stereocenters and the use of various substrates beyond N-allyl amides, demonstrating the value of the chiral heterocycles as strategic intermediates for the synthesis of other enantioenriched target structures.

Recent Advances in N-Heterocyclic Carbene Catalyzed Oxidative Cyclization for the Formation of Heterocycles

T. Pavithra et al.

Summary: Oxidative N-heterocyclic carbene-catalyzed cyclization is an efficient strategy for constructing various N- and O-heterocycles with excellent enantioselectivities. The reaction proceeds via an acyl azolium intermediate, which is an active species capable of forming a diverse array of heterocycles through reactions with suitable substrates. Recent reports have summarized different strategies for formal annulations between incoming substrates and acyl azolium or other active intermediates, leading to the formation of various heterocycles.
