Early and late stage gene therapy interventions for inherited retinal degenerations

Catherine Botto et al.

Summary: Inherited and age-related retinal degeneration are major causes of untreatable blindness. Genetic factors play a significant role in these diseases, and gene therapy is rapidly becoming a viable treatment approach. This article provides an overview of retinal gene therapy methods and their relevance to patients with inherited retinal degenerations.

Control of Microbial Opsin Expression in Stem Cell Derived Cones for Improved Outcomes in Cell Therapy

Marcela Garita-Hernandez et al.

Summary: Human-induced pluripotent stem cell-derived organoids are crucial for modeling human organ development and diseases, with cell-type-specific promoters playing a significant role in controlling gene expression and improving cell sorting efficiency. Use of a 1.7kb L-opsin promoter has shown enhanced gene expression and therapeutic outcomes in cell therapy settings. Promoter activity is essential for regulating transgene expression during the maturation of hiPSC retinal derivatives.

Low Immunogenicity and Immunosuppressive Properties of Human ESC- and iPSC-Derived Retinas

Suguru Yamasaki et al.

Summary: Research has shown that hESC- and iPSC-derived retinas express low levels of HLA-I and minimal HLA-II in vitro, possess the ability to suppress immune activation of lymphocytes in co-culture, and partially suppress activated immune cells via the transforming growth factor β signaling pathway. These findings support the potential use of allogeneic hESC- and iPSC-derived retinal transplantation in future clinical applications.

Immunologic Rejection of Transplanted Retinal Pigmented Epithelium: Mechanisms and Strategies for Prevention

Carson C. Petrash et al.

Summary: The potential of replacing dysfunctional retinal pigmented epithelium (RPE) with stem cell grafts to improve vision in patients with retinal disorders is being explored by numerous research groups. However, factors such as immune responses, unique immunologic environments, and variability in assessment techniques and drug regimens complicate comparisons of findings and determination of successful outcomes in RPE transplant trials. Further research is needed to address these complicating factors and improve the design of future transplants and investigations.

Ultrathin micromolded 3D scaffolds for high-density photoreceptor layer reconstruction

In-Kyu Lee et al.

Summary: Polymeric scaffolds are being used in therapeutics for blinding disorders affecting the outer retina, with recent advances in engineering producing optimized structures for retinal pigment epithelium delivery. A new biodegradable scaffold patterned for high-density photoreceptor replacement has been developed, which may have applications in multicellular layers for outer retinal reconstruction and other tissues. This approach could also be useful for producing various micro- and nanoscale structures for multilayered cell delivery.

Summary: Age-related macular degeneration (AMD) is a prevalent irreversible impairment in the elderly population, and stem cell therapies are being considered as a potentially viable treatment option. Clinical studies and applications focusing on pluripotent stem cell-derived retinal pigment epithelial (RPE) cells are progressing, while controversy surrounds the use of somatic stem cells in clinical trials. The field of retinal regenerative medicine using stem cells is expected to advance steadily with the incorporation of new technologies from various fields, emphasizing the importance of ophthalmologists in this area.

The effect of retinal scaffold modulus on performance during surgical handling

Rion J. Wendland et al.

Summary: The study investigated the effects of scaffold modulus on surgical handling using photocrosslinked poly(ethylene glycol) dimethacrylate as a model polymer. It found that scaffolds with shear modulus above 35 kPa performed satisfactorily in handling, while those below this limit were too fragile for reliable transplantation. Moreover, there was increased variability in surgical handling performance in samples with shear moduli above 800 kPa.
