Androgen Reduces Mitochondrial Respiration in Mouse Brown Adipocytes: A Model for Disordered Energy Balance in Polycystic Ovary Syndrome

Avi Lerner et al.

Summary: Polycystic ovary syndrome (PCOS) is associated with reduced postprandial thermogenesis and abnormalities in adipose tissue function, with excess androgen inhibiting brown adipogenesis, attenuating thermogenic activation, and reducing mitochondrial respiration in brown adipose tissue (BAT). These findings may contribute to the reduced postprandial thermogenesis and increased tendency to obesity observed in women with PCOS.

Reactive Oxygen Species: Beyond Their Reactive Behavior

Arnaud Tauffenberger et al.

Summary: Cellular homeostasis is crucial for organism development and aging, with disruption leading to health degradation and death. Reactive oxygen species (ROS) are strongly linked to health decline and neurological disorders, disrupting macromolecules and playing complex roles in cell signaling, particularly in the brain where they impact neuronal survival and synaptic plasticity.

The Mitochondrial Permeability Transition: Nexus of Aging, Disease and Longevity

Hagai Rottenberg et al.

Summary: mPTP activity is enhanced in aging and aging-driven degenerative diseases, releasing harmful substances that accelerate cell aging and disease progression. Autophagy, a protective mechanism, depends on the inhibition of mPTP for its effectiveness. Inhibition of autophagy and mitophagy in aging cells leads to increased mPTP activation, contributing to neurodegenerative diseases. Enhancing autophagy and inhibiting mPTP activation through drugs and lifestyle modifications can promote longevity and healthspan.

The Role of Mitochondrial Adaptation and Metabolic Flexibility in the Pathophysiology of Obesity and Insulin Resistance: an Updated Overview

Dimitrios Tsilingiris et al.

Summary: Metabolic flexibility refers to the dynamic responses of cellular oxidative machinery to changes in energy substrate availability, and its gradual loss is implicated in the development of obesity-related complications. Mitochondria, as crucial organelles in energy metabolism, play a key role in adaptive responses to environmental challenges and are seen as functional components of metabolic flexibility. There is concrete evidence supporting impaired mitochondrial adaptation as a main component of systemic metabolic inflexibility in obesity-related conditions, although causality issues and generalizability of findings remain inconclusive.

Mitochondrial Dynamics and Mitophagy in Skeletal Muscle Health and Aging

Jean-Philippe Leduc-Gaudet et al.

Summary: The maintenance of mitochondrial integrity is crucial for muscle health, with dysfunctional mitochondrial dynamics and mitophagy potentially contributing to sarcopenia. Enhancing mitophagy in skeletal muscle may be a promising therapeutic target to prevent or treat skeletal muscle dysfunction in the elderly.

Mammalian Target of Rapamycin Signaling Pathway Regulates Mitochondrial Quality Control of Brown Adipocytes in Mice

Bahetiyaer Huwatibieke et al.

Summary: The study demonstrates that the mTOR signaling pathway affects the transformation of adipocytes by regulating mitochondrial quality control in brown adipocytes. Activation of mTOR signaling inhibits the mitochondrial quality control and promotes the transformation from brown to white adipocytes, while inhibition of mTOR signaling enhances mitochondrial quality control and inhibits the phenotypic shift.

Mechanism of insulin resistance in obesity: a role of ATP

Jianping Ye

Summary: This review suggests that ATP may serve as the primary signal for insulin resistance, as elevated ATP production in insulin-sensitive cells under obese conditions contributes to systemic insulin resistance through various mechanisms. The prevention of ATP overproduction is proposed as a key strategy for insulin sensitization in the cellular protective response to substrate oversupply.

Summary: FOXO proteins are transcription factors that are involved in regulating stress responses to maintain cellular homeostasis. Dysregulation of FOXO protein functions contributes to cardiovascular disease, cancer, diabetes, and neurological diseases. This review summarizes the biology of FOXO proteins, their roles in disease and longevity, and discusses pharmacological approaches to develop FOXO-targeting therapeutics.
