Three-Dimensional Primary Cell Culture: A Novel Preclinical Model for Pancreatic Neuroendocrine Tumors

Simon Leonhard April-Monn et al.

Summary: This study established a novel in vitro 3-dimensional human primary PanNET culture system as a preclinical model for personalized therapy selection. The platform allows multicenter sample collection and drug screening, showing that primary cells isolated from PanNET patients can form islet-like tumoroids in vitro and display varying therapeutic response profiles. The model is believed to be suitable for personalized drug testing in a larger patient cohort, aiding in identifying novel markers predicting treatment response and refining PanNET therapy.

The Role of Stellate Cells in Pancreatic Ductal Adenocarcinoma: Targeting Perspectives

Yang Wu et al.

Summary: PDAC progression is influenced by PSCs, which through interactions with PCCs and other stroma cells, establish a tumor microenvironment that promotes tumor growth, metastasis, and chemoresistance. Targeting stroma has emerged as a promising strategy for PDAC therapy, with several novel strategies proposed to intervene in this complex crosstalk.

Advances in biofabrication techniques for collagen-based 3D in vitro culture models for breast cancer research

John Redmond et al.

Summary: Collagen is an ideal biomaterial for cell culture due to its abundance in the ECM, and recent advances have shown promise for its use as 3D culture platforms for breast cancer research. While 2D in vitro culture models have limitations, 3D models aiming to replicate the tumor microenvironment have the potential to provide more accurate insights into cancer development and treatment.

Epidemiology, Incidence, and Prevalence of Neuroendocrine Neoplasms: Are There Global Differences?

Satya Das et al.

Summary: The purpose of the review is to explore global epidemiologic trends of gastroenteropancreatic neuroendocrine tumors (NETs), looking at differences in incidence, prevalence, distribution, and overall survival between different regions. GEP NET incidence rates are rising steadily in North America, Asia, and Europe, with differences in distribution of NETs among regions. Improved overall survival for GEP NET patients is also observed over time.

Extracellular Vesicle Mediated Tumor-Stromal Crosstalk Within an Engineered Lung Cancer Model

Kayla F. Goliwas et al.

Summary: This study developed three-dimensional lung tumor models to mimic the human tumor microenvironment, demonstrating interactions between tumors and stroma mediated by extracellular vesicles (EVs) and their influence on immunomodulation. Inhibiting immune checkpoints in a 3D model including T cells showed suppression of tumor growth and reduction of hypoxia within the tumor microenvironment.

3D Cancer Models: Depicting Cellular Crosstalk within the Tumour Microenvironment

Teresa Franchi-Mendes et al.

Summary: The tumour microenvironment consists of multiple non-cancerous cells that interact with tumour cells, influencing disease progression and therapeutic responses. In vitro 3D models have been developed to mimic the complexity of the tumour microenvironment, providing insights for cancer research and drug discovery. Understanding the role of each cell type in the microenvironment is crucial for uncovering potential therapeutic targets.

Prognostic Implications of the Immune Tumor Microenvironment in Patients With Pancreatic and Gastrointestinal Neuroendocrine Tumors

Marina Baretti et al.

Summary: This study aimed to characterize the tumor microenvironment in gastroenteropancreatic neuroendocrine tumors and its association with progression-free survival (PFS). Patients with shorter PFS tended to have larger tumors, mainly in the pancreas, while those with longer PFS had higher intratumoral CD3(+) T-cell infiltrates, potentially contributing to their longer PFS after resection.
