Empty Sella Syndrome Associated with Growth Hormone Deficiency: the First Case Report of Weiss-Kruszka Syndrome

Summary: Weiss-Kruszka syndrome (WSKA) caused by heterozygous loss-of-function variants in ZNF462 gene is an extremely rare genetic disorder characterized by craniofacial features and neurodevelopmental impairment, which may also present with additional clinical manifestations such as growth hormone deficiency. The identified novel de novo heterozygous nonsense variant c.4185del; p.(Met1396Ter) in ZNF462 is associated with primary empty sella syndrome (ESS) and GHD in the reported case. Further clinical and functional studies are required to understand this association better.
