Screening and characterization of a diverse panel of metagenomic imine reductases for biocatalytic reductive amination

James R. Marshall et al.

Summary: Over 300 new imine reductases have been discovered in this study, and a large and diverse panel of 384 enzymes has been produced for screening. A high-throughput screening method has also been developed. Through these methods, imine reductase biocatalysts capable of accepting structurally demanding ketones and amines for excellent synthesis have been identified.

Engineering Thermostability in Artificial Metalloenzymes to Increase Catalytic Activity

Megan Doble et al.

Summary: Protein engineering has been used to improve the thermostability of an artificial metalloenzyme, resulting in enhanced activity in high-temperature reactions. The modified metalloenzymes showed a significant increase in turnover number during the hydroformylation of 1-octene, while maintaining a high selectivity for linear aldehyde above 80%.

Chemogenetic Evolution of a Peroxidase-like Artificial Metalloenzyme

Ulrich Markel et al.

Summary: Directed evolution has been successful in enhancing enzyme engineering. In this study, an ArM with peroxidase-like properties was designed and evolved using the nitrobindin variant, NB4. Key to boosting peroxidase activity was a substitution to arginine in the distal cofactor environment. The ArM showed high efficiency in decolorization of dyes and could be recycled multiple times.

An artificial ruthenium-containing β-barrel protein for alkene-alkyne coupling reaction

Andreas Thiel et al.

Summary: A modified Cp*Ru complex with a maleimide group was covalently attached to a cysteine of an engineered variant of Ferric hydroxamate uptake protein component: A (FhuA). This synthetic metalloprotein catalyzed the intermolecular alkene-alkyne coupling reaction. Compared to the protein-free Cp*Ru catalyst, the biohybrid catalyst showed higher regioselectivity in producing the linear product.
