SHMT inhibition is effective and synergizes with methotrexate in T-cell acute lymphoblastic leukemia

Juan C. Garcia-Canaveras et al.

Summary: Folate metabolism is crucial for cell growth, with the enzyme SHMT playing a key role in providing 1C units for nucleotide biosynthesis. Inhibition of SHMT by SHIN2 shows promise in improving survival in T-ALL, especially when used in combination with methotrexate.

The Source of Glycolytic Intermediates in Mammalian Tissues

Tara TeSlaa et al.

Summary: Glycolysis is central to organismal metabolism, with circulating glucose and intracellular glycogen being the main sources of glycolytic intermediates. Gluconeogenesis also contributes to glycolysis to a lesser extent. Different tissues show variations in utilizing circulating glucose for glycolysis, with most tissues relying on glycogen breakdown for glycolytic intermediates. Overall, the body slowly breaks down internal glycogen stores while select tissues rapidly catabolize circulating glucose for oxidation.

In Vivo Evidence for Serine Biosynthesis-Defined Sensitivity of Lung Metastasis, but Not of Primary Breast Tumors, to mTORC1 Inhibition

Gianmarco Rinaldi et al.

Summary: This study found that there are different metabolic and nutrient requirements to activate growth signaling between lung metastatic niche and primary breast cancer site. Breast cancer-derived lung metastases use the serine biosynthesis pathway to fuel mTORC1 growth signaling, while primary breast tumors do not rely on this pathway.

Serine synthesis pathway inhibition cooperates with dietary serine and glycine limitation for cancer therapy

Mylene Tajan et al.

Summary: The study demonstrates that inhibition of PHGDH in the serine synthesis pathway, in conjunction with serine and glycine depletion, can inhibit one-carbon metabolism and cancer growth to enhance therapeutic efficacy. This treatment shows effectiveness against drug-resistant tumors in mice, although there are differences in the ATF4 response between in vitro and in vivo settings.

Isotope tracing in adult zebrafish reveals alanine cycling between melanoma and liver

Fuad J. Naser et al.

Summary: The study revealed that melanoma consumes significantly more glucose than other tissues and the tumor liver alanine cycle plays a crucial role in maintaining circulating glucose levels in diseased animals. Inhibition of this cycle reduced tumor burden, highlighting the importance of metabolic crosstalk between tumors and distant tissues.

Cell-programmed nutrient partitioning in the tumour microenvironment

Bradley Reinfeld et al.

Summary: The study found that in the tumor microenvironment, different cell types have varying abilities to access and consume glucose and glutamine, with nutrient partitioning being programmed in a cell-intrinsic manner through mTORC1 signaling and the expression of genes related to glucose and glutamine metabolism.

Local production of lactate, ribose phosphate, and amino acids by human triple-negative breast cancer

Jonathan M. Ghergurovich et al.

Summary: The study explored the metabolism of human triple-negative breast cancer and found that the tumor predominantly ferments glucose to lactate, with most ribose phosphate still being produced by oxPPP. Glucose is also utilized for the synthesis of various amino acids, with pyruvate and lactate labeling in tumors exceeding that found in serum. This indicates that lactate exchange via monocarboxylic transporters is less prevalent in human TNBC compared to normal tissues or non-small cell lung cancer.
