DNA methylation alterations and their potential influence on macrophage in periodontitis

Yiyang Jiang et al.

Summary: The methylation alterations of certain genes in periodontitis can cause abnormal expression of proteins, leading to changes in the local microenvironment and affecting the activation states of immune cells, including macrophages. This has implications for the host immune response and may contribute to differential susceptibility and treatment outcomes in periodontitis.

The neutrophil subset defined by CD177 expression is preferentially recruited to gingival crevicular fluid in periodontitis

Agnes Dahlstrand Rudin et al.

Summary: Research has shown that the CD177(+)neutrophil subset is preferentially recruited to the gingival crevice of periodontitis patients, indicating its potential importance in situations of microbe-driven inflammation.

Periodontal Inflammation Primes the Systemic Innate Immune Response

N. Fine et al.

Summary: The presence of periodontal diseases is closely related to other chronic inflammatory conditions such as cardiovascular disease, diabetes, and arthritis. Studies show that periodontal tissue inflammation affects the production and activation of polymorphonuclear neutrophils (PMNs), contributing to a hyperinflammatory response in the body. This systemic effect of PD on immune response can exacerbate inflammatory reactions when faced with secondary infections, highlighting the complex interactions between PD and other inflammatory diseases.

LDGs versus NDGs in patients with oral squamous cell carcinoma (OSCC)

Marzena Garley et al.

Summary: This study identified the presence of two populations of human neutrophils in patients with oral squamous cell carcinoma (OSCC): low-density neutrophils (LDNs) and normal-density neutrophils (NDNs). A higher percentage of LDNs and Th17 was observed while the percentage of NDNs was lower in these patients compared to the control group. The neutrophil to lymphocyte ratio (NLR) was elevated in cancer patients, with the highest IL-17 expression found in the LDNs population. However, no influence of IL-17 on squamous cell carcinoma proliferation was determined. These findings suggest a potential reference point for new therapies and diagnostic/prognostic value of LDNs/NDNs and NLR in OSCC patients.

TGF-β1 and IL-17A comediate the protumor phenotype of neutrophils to regulate the epithelial-mesenchymal transition in oral squamous cell carcinoma

Tao Yu et al.

Summary: This study investigated the role of TGF-beta 1 and IL-17A in modulating neutrophil function in oral squamous cell carcinoma (OSCC). The findings suggest that the cooperative effect of these two factors promotes a protumorigenic phenotype in neutrophils, leading to enhanced progression of OSCC cells. Additionally, specific neutrophil-associated markers in peripheral blood of OSCC patients may serve as predictive indicators for disease progression.

Summary: The prevalence of periodontal disease increases with age, likely due to systemic inflammatory dysregulation. Age-related changes to macrophages may have a detrimental effect on disease recovery in aging populations, contributing to the pathogenesis of periodontal disease.

Local and systemic mechanisms linking periodontal disease and inflammatory comorbidities

George Hajishengallis et al.

Summary: Periodontitis is causally linked to the development of other chronic inflammatory diseases outside the oral mucosa, and intervention studies have shown that local treatment of periodontitis can ameliorate surrogate markers of comorbid conditions. Experimental animal studies have further strengthened the potential causal link by establishing biologically plausible and clinically consistent mechanisms. Understanding the impact of disseminated periodontal pathogens and systemic inflammation associated with periodontitis on extra-oral pathologies may lead to new therapeutic options to reduce the risk of periodontitis-associated comorbidities.
