Mechanisms of Environment-Induced Autoimmunity

Summary: While many environmental exposures are suggested as triggers for preclinical autoimmunity, only a few have been confidently linked to autoimmune diseases. Lung is a common site where chronic exposure can result in cellular toxicity, tissue damage, inflammation, and fibrosis, exacerbated by particulate material. The process involves posttranslational modification of self-antigens, formation of tertiary lymphoid structures containing B cells, and generation of autoantibodies, which may have diagnostic specificity under gene-environment interactions. Understanding the molecular and cellular requirements distinguishing preclinical autoimmunity from clinical autoimmune disease could help determine the significance of environmental exposures in human autoimmune disease.
