Curation and expansion of Human Phenotype Ontology for defined groups of inborn errors of immunity

Summary: This study aims to expand the Human Phenotype Ontology (HPO) for the depiction of inborn errors of immunity (IEIs) and reannotate IEIs with HPO terms, providing accurate and standardized phenotypic descriptions. By expert review and machine learning, the branches of HPO were expanded and IEI diseases were reannotated with HPO terms. This allows for improved computational matching of IEI cases to their known diagnosis and enhanced phenotype-driven disease classification.

National external quality assessment for next-generation sequencing-based diagnostics of primary immunodeficiencies

Comparison of the diagnostic yield of aCGH and genome-wide sequencing across different neurodevelopmental disorders

Summary: Most consensus recommendations for the genetic diagnosis of neurodevelopmental disorders (NDDs) do not include the use of next generation sequencing (NGS) and are still based on chromosomal microarrays, such as comparative genomic hybridization array (aCGH). This study compares the diagnostic yield obtained by aCGH and clinical exome sequencing in NDD globally and its spectrum of disorders. Clinical exome sequencing showed superior diagnostic yield compared to aCGH in most cases, suggesting that NGS could be used as a first-tier test in the diagnostic algorithm of all NDDs.
