Punicalagin ameliorates collagen-induced arthritis by downregulating M1 macrophage and pyroptosis via NF-kappa B signaling pathway

Gaoran Ge et al.

Summary: The study revealed that punicalagin could ameliorate pathological inflammation in rheumatoid arthritis by modulating macrophage polarization and inhibiting pyroptosis, suggesting its great potential as a therapeutic treatment for human RA.

Modulation of TNFR 1-triggered two opposing signals for inflammation and apoptosis via RIPK 1 disruption by geldanamycin in rheumatoid arthritis

Yukihiko Saeki et al.

Summary: Geldanamycin disrupts RIPK1 and selectively inhibits the TNFR1-triggered NF-kappa B activation signaling pathway, while enhancing the apoptosis signaling pathway upon TNF alpha stimulation, thereby redressing the balance between these two opposing signals in a human rheumatoid synovial cell line. Therapeutic targeting RIPK1 may have great potential for a more fundamental, effective, and safer TNF inhibitor.

Effects of TNF-α on autophagy of rheumatoid arthritis fibroblast-like synoviocytes and regulation of the NF-κB signaling pathway

Yu Wang et al.

Summary: This study found that TNF-α can activate autophagy in FLSs, and inhibition of autophagy can improve the imbalance of proliferation/apoptosis exacerbated by TNF-α, thereby delaying the progression of RA. The NF-kappa B signal pathway may be involved in this regulation.

Summary: The study demonstrated that tangeretin and 5-hydroxy-6,7,8,3',4'-pentamethoxyflavone prevented the progression of rheumatoid arthritis by inhibiting autophagy in synovial cells via the ROS-AKT/mTOR signaling axis.

Summary: The study showed that Pterostilbene combined with physical exercise has a significant preventive effect on rheumatoid arthritis, increasing the MaR1 content, reversing the abnormal expression of inflammatory signal pathways, and inhibiting the proliferation and apoptosis of synovial cells through promoting cell autophagy.

Molecular and Cellular Heterogeneity in Rheumatoid Arthritis: Mechanisms and Clinical Implications

Jianan Zhao et al.

Summary: Rheumatoid arthritis displays significant clinical heterogeneity and personalized responsiveness to standard treatments, with understanding individual variation in cellular and molecular mechanisms being crucial for improving patient outcomes. Pathological heterogeneity derived from genetic, molecular, and cellular factors influences precision medicine and personalized treatment approaches for rheumatoid arthritis.

MicroRNA Variants and HLA-miRNA Interactions are Novel Rheumatoid Arthritis Susceptibility Factors

Shicheng Guo et al.

Summary: Genome-wide association studies have identified over 100 genetic risk factors for rheumatoid arthritis, but they can only explain less than 40% of its heritability. To address the missing heritability, novel functional SNPs were identified through genotyping HLA SNPs and common SNPs located in miRNA. The study discovered new SNPs significantly associated with rheumatoid arthritis and demonstrated significant epistasis effects between certain SNPs.

A randomized, placebo-controlled experimental medicine study of RIPK1 inhibitor GSK2982772 in patients with moderate to severe rheumatoid arthritis

Summary: This study evaluated the safety, pharmacokinetics, and preliminary efficacy of GSK2982772, a RIPK1 inhibitor, in patients with moderate to severe rheumatoid arthritis. The results suggest that inhibition of RIPK1 activity at the exposure levels of GSK2982772 evaluated does not lead to significant clinical improvement in RA.
