Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

The war against pancreatic cancer in 2020-advances on all fronts

Tejeshwar Jain et al.

Summary: In 2020, significant progress was made in three major areas of pancreatic ductal adenocarcinoma (PDAC) research: risk factors, therapeutic resistance, and tumor recurrence. The pathophysiology of obesity-mediated PDAC initiation was elucidated, novel stromal mechanisms of therapeutic resistance were unveiled, and the genetic evolution of recurrent PDAC under therapeutic pressures was tracked in human samples.

The RNA methyltransferase NSUN6 suppresses pancreatic cancer development by regulating cell proliferation

Ruimeng Yang et al.

Summary: This study identified NSUN6 as an important factor involved in regulating cell proliferation in pancreatic cancer and highlighted the potential of novel m5C-based clinical modalities as a therapeutic approach in PC patients. NSUN6 expression correlated with clinicopathologic parameters and was found to play a crucial role in cell proliferation and tumor growth in PC. Furthermore, NSUN6 showed good performance in evaluating tumor recurrence and survival among PC patients.

LncRNAs in cancer: Regulatory and therapeutic implications

Arun Nandwani et al.

Summary: Long noncoding RNAs (lncRNAs) play critical roles in regulating cellular processes and maintaining cell function, associated with a range of diseases and potential therapeutic targets. Interaction with chromatin, proteins, or RNA affects cellular proliferation, migration, differentiation, apoptosis, and cell death, making them key players in various cancers.

Predictive value of m5C regulatory gene expression in pancreatic adenocarcinoma

Xiao Yu et al.

Summary: The study identified significant correlations between m5C regulatory gene changes and key genes in PAAD pathogenesis, such as TP53, BRCA1, CDKN2A, and ATM. Low DNMT3A expression was found to be associated with poor prognosis in PAAD patients as well as with ribosomal processing. This relationship was confirmed in an International Cancer Genome Consortium validation dataset.

m5C-Related lncRNAs Predict Overall Survival of Patients and Regulate the Tumor Immune Microenvironment in Lung Adenocarcinoma

Junfan Pan et al.

Summary: The study focused on the role of lncRNAs associated with RNA methylation in the prognosis of patients with lung adenocarcinoma (LUAD), constructing a risk model with prognostic value and validating and analyzing its effectiveness. The results suggest that the risk model based on m5C-related lncRNAs may be a promising prognostic tool for LUAD management, indicating better prognosis in the low-risk group compared to the high-risk group.

Silencing of CASC8 inhibits non-small cell lung cancer cells function and promotes sensitivity to osimertinib via FOXM1

Xizi Jiang et al.

Summary: The study identified CASC8 as a cancer susceptibility gene closely related to lung cancer, with high expression associated with poor prognosis in patients with lung adenocarcinoma. Silencing CASC8 inhibited proliferation, migration, and invasion in non-small cell lung cancer cell lines, and also promoted sensitivity to osimertinib through FOXM1. This pathway may offer new treatment options for lung cancer patients resistant to targeted therapies.
