Immunoinformatics approach for multiepitope vaccine design against structural proteins and ORF1a polyprotein of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2)

Khalid Mohamed Adam

Summary: A thorough immunoinformatics approach was used to identify suitable epitopes for constructing a vaccine against SARS-CoV-2. The final candidate vaccine construct consists of 29 epitopes and has the potential to trigger both humoral and cell-mediated immune responses. Computational validation and molecular modeling were performed to confirm the efficacy of the vaccine candidate.

Linear epitope landscape of the SARS-CoV-2 Spike protein constructed from 1,051 COVID-19 patients

Yang Li et al.

Summary: By analyzing the serum IgG response of 1,051 COVID-19 patients, researchers identified linear epitopes of the SARS-CoV-2 Spike protein, revealing two regions rich in linear epitopes and identifying variable peptide responses associated with disease severity. Antibodies obtained from immunizing mice with linear epitopes did not show significant neutralizing activity against the virus, suggesting the importance of understanding humoral responses for vaccine refinement.

Immunodominant linear B cell epitopes in the spike and membrane proteins of SARS-CoV-2 identified by immunoinformatics prediction and immunoassay

Kanokporn Polyiam et al.

Summary: This study identified three immunodominant linear B cell epitopes in the SARS-CoV-2 spike receptor-binding domain through immunoinformatics prediction and confirmed them with ELISA. Predictions and confirmations of B cell epitopes in spike, membrane, and envelope proteins were also conducted using convalescent sera from COVID-19 infected patients. The findings offer insights into B cell epitopes of SARS-CoV-2 and immunoinformatics methods for B cell epitope prediction, which can enhance SARS-CoV-2 vaccine development against emergent variants.

A guide to vaccinology: from basic principles to new developments

Andrew J. Pollard et al.

Summary: Immunization is vital for public health policy and there are challenges ahead for immunologists in developing new vaccines. Although immunology has not contributed significantly to vaccine development so far, a better understanding of protective immunity is urgently needed to tackle difficult-to-target pathogens.
