Bacillus velezensis stimulates resident rhizosphere Pseudomonas stutzeri for plant health through metabolic interactions

Summary: Trophic interactions between inoculant Bacillus velezensis SQR9 and indigenous Pseudomonas stutzeri in the cucumber rhizosphere showed environmental dependency and involved synthesis of branched-chain amino acids. The synergistic interaction was only evident in nutrient-rich niches like pellicle biofilm. Furthermore, metabolic modeling and profiling demonstrated facilitation among the bacterial strains, promoting plant growth and alleviating salt stress.

Glycoside hydrolase family 5: structural snapshots highlighting the involvement of two conserved residues in catalysis

Summary: This study revealed the GEI structure of an endo-1,4-beta-glucanase of the GH5 family, RBcel1, as well as the structure of the variant RBcel1_E135A in complex with cellotriose, providing insights into the double-displacement mechanism. Structural analysis showed significant movement of the nucleophilic glutamate residue, and enzymatic assays indicated the crucial roles of the acid/base glutamate residue and a conserved tyrosine residue in both glycosylation and deglycosylation steps. The approach used in this study could be applied to other retaining GHs in clan GH-A.

Probing the determinants of the transglycosylation/hydrolysis partition in a retaining α-L-arabinofuranosidase

Summary: The study focused on exploring the active site of a GH51 retaining alpha-L-arabinofuranosidase from Thermobacillus xylanilyticus through analysis of two mutants, F26L and L352M. The results showed that the mutant R69H-N216W-L352M displayed excellent transglycosylation activity by inducing changes in the donor and acceptor subsites. Additionally, the central role played by the conserved R69 residue in affecting the T/H partition was reaffirmed.

Bi et al. showed that a Crh protein from the phytopathogenic fungus Botrytis cinerea acts as a cytoplasmic effector and elicitor of plant defense, leading to reduced sensitivity to the pathogen in plants expressing this gene.
