Autism Spectrum Disorder: Signaling Pathways and Prospective Therapeutic Targets

Juliana Baranova et al.

Summary: ASD is a complex group of neurodevelopmental disorders with no effective drugs developed for treatment. Mutations in core components and dysfunctions in multiple signaling pathways related to neurological development, metabolism, and immune regulation have been identified. Research aims to develop novel biomarkers for early ASD diagnosis and potential therapeutic targets.

Bumetanide for Core Symptoms of Autism Spectrum Disorder (BAMBI): A Single Center, Double-Blinded, Participant-Randomized, Placebo-Controlled, Phase-2 Superiority Trial

Jan J. Sprengers et al.

Summary: This study tested the efficacy of bumetanide in treating core symptoms of autism spectrum disorder, with results showing no superior effect on the primary outcome but suggesting efficacy on repetitive behavior symptoms for a subset of patients. Adverse effects were mainly diuretic effects, with no statistical influence on treatment outcome.

Major motor and gait deficits with sexual dimorphism in a Shank3 mutant mouse model

Emmanuel Matas et al.

Summary: The Shank3 Delta C/Delta C mice exhibited significant impairments in social behavior, stereotyped behavior, and gait, along with decreased Purkinje cells and mGluR5 expression in the cerebellum. On the other hand, Shank3+/Delta C mice only showed impairments in social behavior and reduced mGluR5 expression to a lesser extent. The deficits were more pronounced in male mice compared to females.

Neurexin1α differentially regulates synaptic efficacy within striatal circuits

M. Felicia Davatolhagh et al.

Summary: In the striatal circuitry, dysregulation of Nrxn1α specifically affects synaptic strength onto the indirect pathway SPNs, while thalamic excitatory inputs to DMS exhibit relatively normal excitatory synaptic strength despite changes in synaptic NMDAR content. This suggests that Nrxn1α modulates striatal function in an input- and target-specific manner.

Excitatory neuronal CHD8 in the regulation of neocortical development and sensory-motor behaviors

Hanseul Kweon et al.

Summary: Deletion of the CHD8 gene in mice leads to near-complete elimination of neocortical structures, yet the mice display mostly normal behaviors, including hyperactivity and increased social interaction. These changes are accompanied by thalamic hyperactivity and enhanced somatosensory function, suggesting a critical role of excitatory neuronal CHD8 in neurodevelopment and a compensatory response in CHD8 haploinsufficiency-related macrocephaly.

Social Behavior in Prepubertal Neurexin 1α Deficient Rats: A Model of Neurodevelopmental Disorders

Katherine E. Kight et al.

Summary: Loss-of-function mutations in synaptic protein neurexin1 alpha are linked to neurodevelopmental disorders like ASD, schizophrenia, and ADHD, affecting social cognition. Using rats as models, the study found deficits in ultrasonic vocalizations and social play in male rats with Nrxn1 alpha deficiency, while nurturing behavior was enhanced in rats with biallelic mutation. Severe deficits in prosocial helping behavior and learning were seen in rats with biallelic mutation, supporting the rat model for Nrxn1 alpha-related disorders.

Restoring glutamate receptosome dynamics at synapses rescues autism-like deficits in Shank3-deficient mice

Enora Moutin et al.

Summary: Mutations in the Shank3 gene lead to autism spectrum disorders through disrupting the integrity of the glutamate receptosome, affecting synaptic plasticity and behaviors. Fine-tuning protein-protein interactions within the glutamate receptosome controlled by changes in neuronal activity play a crucial role in regulating plasticity. Restoring the integrity of the glutamate receptosome and its sensitivity to neuronal activity could rescue synaptic transmission, plasticity, and behaviors associated with Shank3 mutations.

Clara A. Amegandjin et al.

Summary: Regulators of mTORC1, such as Tsc1, play a significant role in neurodevelopmental disorders associated with autism, intellectual disabilities, and epilepsy. The mTORC1 signaling pathway is essential for regulating and maintaining cortical parvalbumin cell connectivity and social behavior.

Summary: Mutations in CNTNAP2 have been linked to a specific form of Autism Spectrum Disorder. Using forebrain organoids derived from induced pluripotent stem cells of patients with this mutation, the study found that the increased volume and cell number were driven by abnormal cellular proliferation and neurogenesis, offering insights into potential therapeutic strategies for ASD.
