Rheumatoid arthritis, systemic lupus erythematosus and primary Sjogren's syndrome shared megakaryocyte expansion in peripheral blood

Yukai Wang et al.

Summary: Rheumatoid arthritis (RA), systemic lupus erythematosus (SLE) and primary Sjogren's syndrome (pSS) exhibit common transcriptional profiles and immune cell compositions in peripheral blood, particularly related to nucleosome assembly and haemostasis. Distinct transcription factor ensembles and gene regulatory networks mainly enrich in hematopoiesis. Upregulated line-specific TFs such as PBX1, GATA1, TAL1, and GFI1B demonstrate a strong gene expression signature of megakaryocyte expansion.

Regulation of Synovial Inflammation and Tissue Destruction by Guanylate Binding Protein 5 in Synovial Fibroblasts From Patients With Rheumatoid Arthritis and Rats With Adjuvant-Induced Arthritis

Mahamudul Haque et al.

Summary: The study found that GBP-5 is up-regulated in RA synovial tissue and joints of rats with AIA, and is induced by IL-1 beta, TNF, and/or IFN gamma in human RASFs. GBP-5 has the potential to restore cellular homeostasis and blunt inflammation and tissue destruction in RA by regulating cytokine signaling pathways.

Local and systemic effects of IL-17 in joint inflammation: a historical perspective from discovery to targeting

Pierre Miossec

Summary: The role of IL-17 in inflammatory and autoimmune diseases has been well established, leading to the development of three registered anti-IL-17 therapies. Ongoing research aims to identify more treatment options and patients who will respond positively, with inhibitors for IL-17 and Th17 cells actively in development.

Elevated serum TREM-1 is associated with periodontitis and disease activity in rheumatoid arthritis

Nevsun Inanc et al.

Summary: TREM-1 and PGLYRP1 levels were higher in RA patients compared to BD and healthy controls, and were associated with periodontitis. In RA patients receiving methotrexate, TREM-1 levels were lower. TREM-1 correlated with CRP, DAS-28, and erythrocyte sedimentation rate, while PGLYRP1 positively correlated with CRP. RA patients had significantly higher salivary bacterial DNA counts than healthy controls. Increased serum TREM-1 levels were associated with PGLYRP1, CRP, and DAS-28-ESR in RA patients with periodontitis.

Interleukin-17A Interweaves the Skeletal and Immune Systems

Mengjia Tang et al.

Summary: The interaction between the immune and skeletal systems is crucial for maintaining skeletal homeostasis, with IL-17A playing a key role; IL-17A has dual effects on osteoclasts and osteoblasts, and is involved in immune-mediated bone diseases; Understanding the pathophysiological roles of IL-17A may offer potential strategies for treating immune-mediated bone diseases.

Circulating and Synovial Pentraxin-3 (PTX3) Expression Levels Correlate With Rheumatoid Arthritis Severity and Tissue Infiltration Independently of Conventional Treatments Response

Marie-Astrid Boutet et al.

Summary: The study found that circulating levels of PTX3 are associated with disease activity and structural damages in early RA patients, while synovial PTX3 is linked to immune cell infiltration and ectopic lymphoid structures in the joint. High levels of PTX3 in the synovium and peripheral blood are associated with chronic inflammation typical of RA, with further mechanistic links requiring additional research.
