Chemokine Receptor 2-targeted Molecular Imaging in Pulmonary Fibrosis A Clinical Trial

Steven L. Brody et al.

Summary: The Cu-64-DOTA-ECL1i PET uptake correlated with CCR2(+) cell infiltration in the lung associated with fibrosis, with inhibition of fibrosis by IL1 beta blockade or pirfenidone reducing CCR2(+) macrophage accumulation. CCR2(+) cells concentrated in perifibrotic regions in lung tissues from patients with IPF, exhibiting intensive signals in fibrotic zones compared to healthy volunteers. These findings suggest a role for imaging CCR2(+) cells within the fibrogenic niche in IPF for personalized therapy and monitoring.

Systematic Review and Meta-analysis of Pirfenidone, Nintedanib, and Pamrevlumab for the Treatment of Idiopathic Pulmonary Fibrosis

Enrica Di Martino et al.

Summary: A systematic review and meta-analysis of three drugs for idiopathic pulmonary fibrosis (IPF) showed that pamrevlumab was more effective in slowing the decline in forced vital capacity (FVC) compared to pirfenidone and nintedanib. Pamrevlumab may be a potential treatment for IPF in phase 3 trials.

Endothelial GATA4 controls liver fibrosis and regeneration by preventing a pathogenic switch in angiocrine signaling

Manuel Winkler et al.

Summary: The study reveals that endothelial GATA4 plays a protective role against perisinusoidal liver fibrosis by repressing MYC activation and profibrotic angiocrine signaling. Therapies targeting the GATA4/MYC/PDGFB/PDGFRb axis offer a promising strategy for prevention and treatment of liver fibrosis.

Summary: This study reveals a novel mechanism by which pulmonary vascular endothelial cells contribute to the pathogenesis of pulmonary fibrosis by regulating the transdifferentiation of microvascular pericytes into myofibroblasts. Targeting myofibroblast formation may prevent the development of pulmonary fibrosis, offering promising therapeutic targets for the disease.

The role of vascular niche and endothelial cells in organogenesis and regeneration

Domenico Ribatti et al.

Summary: The vascular niche refers to the physical and biochemical environment around blood vessels where different cell types can develop and regenerate. Endothelial cells play a crucial role in organ maturation, and dynamic interactions between specific organ endothelial cells and different cellular components are essential for organ morphogenesis and function. Organs, on the other hand, provide cues that shape the structure of the vascular network.

TGF-β as a driver of fibrosis: physiological roles and therapeutic opportunities

Erine H. Budi et al.

Summary: This review focuses on the fibrogenic roles of increased TGF-beta activities and underlying signaling mechanisms in chronic diseases marked by fibrosis. Insight into these roles and mechanisms of TGF-beta as a universal driver of fibrosis has stimulated the development of therapeutic interventions to attenuate fibrosis progression, based on interference with TGF-beta signaling. The promise of these interventions in preclinical and clinical settings will be discussed.

Therapeutic Potential of Quercetin to Alleviate Endothelial Dysfunction in Age-Related Cardiovascular Diseases

Olina Dagher et al.

Summary: The vascular endothelium plays crucial roles in maintaining cardiovascular homeostasis, regulating vascular tone, and processing biochemical substances. Dysfunction of endothelial cells may lead to chronic age-related cardiovascular diseases such as ischemic heart disease. Studies have shown that flavonoid compound quercetin, found in various food sources, may help mitigate the risk factors of endothelial dysfunction such as atherosclerosis and hypertension.
