The protein corona modulates the inflammation inhibition by cationic nanoparticles via cell-free DNA scavenging

Summary: A central paradigm in nanomedicine is that the protein corona surrounding synthetic nanoparticles plays a crucial role in their biological functions. This study focused on evaluating the effects of the protein corona on cationic nanoparticles used as rheumatoid arthritis drugs. The researchers found that hydroxylated nanoparticles exhibited prolonged retention in inflamed joints and demonstrated stronger anti-inflammatory effects. Proteomic analysis revealed that the composition of the protein corona influenced the biodistribution profiles of the nanoparticles.

Controlling cancer-induced inflammation with a nucleic acid scavenger prevents lung metastasis in murine models of breast cancer

Summary: The study demonstrates that PAMAM-G3 can scavenge NA DAMPs and mitigate their proinflammatory effects, reducing lung metastasis. Treatment with PAMAM-G3 decreases cell-free DNA levels and lung metastasis, associated with reduction in inflammatory immune cell subsets and proinflammatory cytokine levels in the tumor and the periphery. The findings support the potential inclusion of NAS therapy in breast cancer patients undergoing standard-of-care surgery.

Topical cationic hairy particles targeting cell free DNA in dermis enhance treatment of psoriasis

Summary: The study reveals that poly(2-(dimethylamino)ethyl methacrylate) (PDMA) grafted hairy silica particles (cSPs) with tunable PDMA length and particle size can effectively scavenge cfDNA in the dermis, treating psoriasis. Among them, cSPs of 700 nm diameter show excellent performance with more accumulation and longer retention in psoriatic lesions, leading to better treatment results at a lower administration frequency.
