Reprogramming of glucose metabolism of cumulus cells and oocytes and its therapeutic significance

Shogo Imanaka et al.

Summary: This review summarizes the molecular mechanisms of glucose metabolism during oocyte maturation, focusing on the role of pyruvate dehydrogenase (PDH) and the metabolic balance between aerobic glycolysis and the TCA cycle/OXPHOS. The understanding of the PDK-PDH axis is crucial for developing novel therapeutic strategies to improve oocyte quality.

Reversing tozasertib resistance in glioma through inhibition of pyruvate dehydrogenase kinases

Esther P. Jane et al.

Summary: This study demonstrates that targeting aurora kinases with tozasertib can effectively inhibit cell growth and migration in high-grade glioma cell lines, leading to cell death. However, sustained exposure to tozasertib can result in drug resistance, with upregulation of PDK genes playing a crucial role in the emergence of resistance.

Acrylamide inhibits autophagy, induces apoptosis and alters cellular metabolic profiles

Dan Song et al.

Summary: ACR exposure has been shown to cause neurotoxicity, reproductive toxicity, and carcinogenicity, but the exact mechanism of its toxicity remains unknown. In this study, it was demonstrated that ACR inhibited autophagic flux, increased ROS content, and led to apoptosis and inflammatory responses. Metabolomic analysis revealed significant differences in metabolic profiles between ACR treated and control cells.

Anti-Warburg Effect of Melatonin: A Proposed Mechanism to Explain its Inhibition of Multiple Diseases

Russel J. Reiter et al.

Summary: Glucose is a crucial nutrient for cells, but in some pathological cells, aerobic glycolysis may occur due to the inhibition of pyruvate dehydrogenase complex. Glycolytics like melatonin can convert diseased cells to a healthier phenotype by inhibiting hypoxia-inducible factor-1 alpha and promoting intramitochondrial conversion of pyruvate into acetyl coenzyme A, thus explaining its actions in inhibiting various diseases.

Summary: Metabolic alterations in cancer tissues show increased activity in enzymes related to glycolysis, pentose phosphate pathway (PPP), and fatty acid synthesis, while the tricarboxylic acid (TCA) cycle and beta-oxidation are suppressed. These changes are consistent with metabolomics results, with a similar pattern observed in noncancer tissues as liver disease progresses.

Metallothionein Attenuated Arsenic-Induced Cytotoxicity: The Underlying Mechanism Reflected by Metabolomics and Lipidomics

Zihe Qi et al.

Summary: Arsenic ions (As3+) are recognized as a hazard to human health, but metallothionein (MT) rich in cysteine can alleviate As3+-induced cytotoxicity. As3+ disrupts phospholipids metabolism, induces cell membrane damage, and perturbs energy and amino acid metabolism. MT rescues As3+-induced metabolic disorder and suppresses reactive oxygen species accumulation by forming As3+-MT complex.

Transformation in band energetics of CuO nanoparticles as a function of solubility and its impact on cellular response

Archini Paruthi et al.

Summary: This study demonstrates that the toxicological potential of CuO nanoparticles is a temporal function of band energies, particularly their overlap with the intracellular redox potential, leading to the release of ionic species in the cytotoxic regime. The dissolution of CuO nanoparticles in different media resulted in physico-chemical transformations and significant changes in bandgap energy. Assessment of intracellular ROS generation and changes in cell viability, apoptosis, and necrosis revealed the biological effects of these nanoparticles. Additionally, mRNA expression analysis of inflammatory cytokines showed a response to nanoparticle treatment within 6 hours.
