Colchicine for the treatment of cardiovascular diseases: old drug, new targets

Alessandro Andreis et al.

Summary: Colchicine has shown emerging potential in the treatment of cardiovascular diseases, targeting key mechanisms of inflammation and adverse ventricular remodelling, making it a valuable, well-tolerated and inexpensive drug in this setting.

Interleukin 1α: a comprehensive review on the role of IL-1α in the pathogenesis and treatment of autoimmune and inflammatory diseases

Giulio Cavalli et al.

Summary: IL-1α is a crucial pro-inflammatory cytokine that is present in almost all cell types and plays a central role in the pathogenesis of various organ or tissue inflammatory diseases. Three biologics are available to target and inhibit IL-1α for therapeutic management of these conditions.

Usefulness of Beta-Blockers to Control Symptoms in Patients With Pericarditis

Massimo Imazio et al.

Summary: This study explored the potential efficacy of beta-blockers in improving symptom control in patients with pericarditis. The results showed that the use of beta-blockers on top of standard anti-inflammatory therapies was associated with improved symptom control, with lower frequency of symptoms persistence at 3 weeks and a trend towards a reduction of recurrences at 18 months.

An update on the pathophysiology of acute and recurrent pericarditis

Aldo Bonaventura et al.

Summary: Recent progress has been made in understanding the pathophysiology of pericarditis. Studies have shown the role of interleukin (IL)-1 and NLRP3 inflammasome in the inflammatory reaction to various stimuli, with viral infections being a common cause. This knowledge could potentially lead to targeted therapies in the future.

Anti-interleukin 1 agents for the treatment of recurrent pericarditis: a systematic review and meta-analysis

Massimo Imazio et al.

Summary: Anti-interleukin-1 (IL-1) agents such as anakinra and rilonacept are effective in preventing recurrences in patients with corticosteroid-dependent and colchicine-resistant recurrent pericarditis (RP) without severe adverse events. However, the use of anti-IL-1 agents is associated with an increased risk of adverse events such as injection-site reactions and infections.
