A Multi-Enzyme Cascade Reaction for the Production of 2′3′-cGAMP

Summary: Multi-enzyme cascade reactions have become increasingly important for synthesizing complex products, offering advantages such as the ability to synthesize complex molecules efficiently without purification of intermediates, easier handling of unstable intermediates, and dealing with unfavorable thermodynamics. In this study, a four-enzyme cascade was successfully developed for the synthesis of 2'3'-cGAMP, achieving comparable synthesis rates to single-step reactions and yielding results on par with chemical synthesis through iterative optimization of substrate, cofactor, and enzyme concentrations. The established enzyme cascade demonstrated the performance capabilities of multi-enzyme cascades for synthesizing pharmaceutically relevant products.
