Standardized microfluidic assessment of red blood cell-mediated microcapillary occlusion: Association with clinical phenotype and hydroxyurea responsiveness in sickle cell disease

Summary: This study introduced a standardized in vitro microfluidic assay and Occlusion Index (OI) for assessing red blood cell (RBC)-mediated microcapillary occlusion and its clinical associations in sickle cell disease (SCD). The results identified two sub-populations within HbSS subjects based on OI distribution, with higher OI subjects showing lower hemoglobin, fetal hemoglobin levels, and mean corpuscular volume, while also being more likely to have concurrent intrapulmonary shunting. Further analysis showed lower OI associated with hydroxyurea responsiveness in HbSS subjects.
