Efficient Screening of Glycan-Specific Aptamers Using a Glycosylated Peptide as a Scaffold

Wei Li et al.

Summary: This study presents a novel approach to efficiently screen aptamers towards specific glycans with complex structures using a glycosylated peptide scaffold. By alternately conducting positive and negative selection steps, aptamers with satisfactory binding abilities to the target glycan A2G2S2 were successfully selected, which were then used for fluorescent imaging of cancerous liver cells to achieve differentiation from normal liver cells.

Integrin αvβ6-TGFβ-SOX4 Pathway Drives Immune Evasion in Triple-Negative Breast Cancer

Archis Bagati et al.

Summary: SOX4 transcription factor plays a key role in mediating resistance to T cell-mediated cytotoxicity in TNBC cells, and inhibiting it can increase the expression of genes in important immune pathways. The integrin αvβ6 receptor regulates the expression of SOX4, and targeting the integrin αvβ6-TGFβ-SOX4 pathway provides therapeutic opportunities for TNBC and other aggressive cancers of epithelial origin.

Cell surface thermal proteome profiling tracks perturbations and drug targets on the plasma membrane

Mathias Kalxdorf et al.

Summary: This study introduces cell surface thermal proteome profiling for comprehensive characterization of protein changes induced by ligands. It reveals drug binding to receptors, remodeling of T cell receptor complexes, and a competition-based method to measure G-protein-coupled receptor antagonist engagement. Remodeling of the plasma membrane proteome in response to a TGFB receptor inhibitor explains the antimetastatic effects of the drug.

Cancer-secreted exosomal miR-21-5p induces angiogenesis and vascular permeability by targeting KRIT1

Qinglian He et al.

Summary: Cancer-secreted exosomes containing miR-21-5p play a crucial role in promoting angiogenesis and vascular permeability in colorectal cancer by suppressing KRIT1 and activating the beta-catenin signaling pathway. The expression levels of miR-21-5p in circulating exosomes are significantly higher in CRC patients compared to healthy donors, suggesting its potential as a therapeutic target for CRC.

Machine learning to guide the use of adjuvant therapies for breast cancer

Ahmed M. Alaa et al.

Summary: The Adjutorium model developed using machine learning technology accurately predicts the survival benefit of early breast cancer patients under different treatment options, demonstrating significantly improved accuracy compared to the current clinical tool PREDICT v2.1 in both internal and external validation. It substantially enhances accuracy in specific subgroups not well-served by existing models and is now implemented as a web-based decision support tool to assist in adjuvant therapy decisions for women with early breast cancer globally.

Neutrophil oxidative stress mediates obesity-associated vascular dysfunction and metastatic transmigration

Sheri A. C. McDowell et al.

Summary: Quail and colleagues demonstrate that obesity leads to impairment of vascular integrity through neutrophil-dependent mechanisms, facilitating breast cancer metastasis to the lungs. Targetable mechanisms underlying cancer progression in the obese population are revealed.
