CRISPR-Cas9 mediated OsMIR168a knockout reveals its pleiotropy in rice

Jianping Zhou et al.

Summary: MicroRNA168 (MIR168) is a crucial miRNA in plants, targeting the RNA-induced silencing complex component Argonaute 1 (AGO1) for regulation of plant growth and environmental stress responses. Through generating clean OsMIR168a deletion mutants using CRISPR-Cas9 strategy, this study found that OsMIR168a plays a significant role in regulating important agronomic traits in rice by targeting genes such as OsAGO1 and OsAGO18. Additionally, differential expression of protein-coding genes and miRNAs in the OsMIR168a mutants suggests a major transcriptional regulatory role for OsMIR168a, particularly through its potential target genes.

Transcriptional and post-transcriptional regulation of heading date in rice

Shirong Zhou et al.

Summary: Rice, as a facultative short day plant, plays a vital role as both a model plant for genetic studies and a staple crop for much of the world's population. Recent research has focused on the transcriptional and post-transcriptional regulation of heading date genes in rice, with an emphasis on post-translational modifications. Efforts are being made to collect genetic resources in a common background to improve breeding strategies for rice cultivars that can adapt to diverse environments.

The transcriptional repressor OsPRR73 links circadian clock and photoperiod pathway to control heading date in rice

Liwen Liang et al.

Summary: The rice floral inhibitor OsPRR73 plays a key role in regulating flowering time by binding to the promoters of floral and circadian clock genes. Its overexpression results in late heading while knockout mutants exhibit early heading in certain light conditions. OsPRR73 is involved in a feedback loop of the rice clock, connecting the photoperiod flowering pathway in rice.

SDC mediates DNA methylation-controlled clock pace by interacting with ZTL in Arabidopsis

Wenwen Tian et al.

Summary: The molecular bases of eukaryotic circadian clocks involve transcriptional-translational feedback loops and epigenetic codes, with DNA methylation playing a significant role in fine-tuning circadian rhythms. Global genome DNA hypomethylation can prolong the circadian period in Arabidopsis, with the SDC protein acting as a link between DNA hypomethylation and a proteolytic cascade to tune the circadian clock.

DNA methylation is involved in acclimation to iron-deficiency in rice (Oryza sativa)

Shuo Sun et al.

Summary: This study identified widespread hypermethylation in rice seedlings under Fe-deficient conditions, particularly in the CHH context. Integrative analysis revealed a negative correlation between transcript abundance of Fe-deficiency-induced genes and nearby TEs, and a positive correlation with 24-nucleotide siRNAs. Both pharmacological and genetic manipulation of DNA methylation demonstrated a crucial role of specific methylation patterns, directed by siRNAs, in acclimation of rice to Fe-deficient conditions. Comparisons with other reports suggest that this may be a universal mechanism for plants to adapt to limited nutrient availability.

DNA demethylases remodel DNA methylation in rice gametes and zygote and are required for reproduction

Shaoli Zhou et al.

Summary: The study revealed that DNA methylation is locally reconfigured after fertilization and intensified during embryogenesis in rice. Three rice DNA glycosylases, DNG702, DNG701, and DNG704, were found to demethylate DNA at distinct genomic regions in gametes and zygotes, playing a crucial role in zygotic gene expression and development. The results suggest that active DNA demethylation in gametes and zygotes is critical for gene expression and reproduction in rice.
