By comparing two recombination strategies, a highly DOX-resistant variant was obtained with significantly improved resistance in various organic solvents due to attracting water molecules by charged surface substitutions. The obtained knowledge and strategies may be applicable for engineering other enzymes with a similar alpha/beta-hydrolase fold.

Less Unfavorable Salt Bridges on the Enzyme Surface Result in More Organic Cosolvent Resistance

Summary: A smart salt bridge design strategy was used to improve the organic solvent resistance and thermostability of Bacillus subtilits Lipase A (BSLA), resulting in variants with significantly enhanced resistance towards organic solvents and improved thermal stability. Molecular dynamics simulations showed that locally refined flexibility and strengthened hydration played key roles in the increased resistance in organic solvents and at elevated temperatures. This salt bridge redesign approach could be a powerful tool for protein engineers to design lipases and other alpha/beta-hydrolases with enhanced resistance to organic solvents and/or thermal stability.

Chemogenetic Evolution of a Peroxidase-like Artificial Metalloenzyme

Summary: This study demonstrated that intrinsic lipase stability and preservation of hydration shells are two synergistic design principles for retaining enzymatic activity in ionic liquids (ILs). By rational design and screening of variants, the resistant enzyme M1a showed significantly improved activity and resistance to various ILs. Computational analysis revealed the molecular basis for the enhanced stability and hydration shells in these variants, providing insights for future design of IL-resistant enzymes.
