Rotaxane Dendrimers: Alliance between Giants

Summary: The conversion of generation 2 and 3 PAMAM dendrimers into N-(2,3-dihydroxy)propyl derivatives, by the addition of enantiomerically pure S- and R-glycidol, resulted in homochiral dendrimers with higher binding ability, especially the R-glycidol derivatives showed increased accumulation in cancer cells. The G3 PAMAM-based dendrimers were more efficiently taken up compared to G2 derivatives, and dendrimers furnished with S- and R-glycidol were highly biocompatible.

A catalysis-driven artificial molecular pump

Summary: This study introduces an autonomous chemically fueled information ratchet that continuously pumps crown ether macrocycles from bulk solution onto a molecular axle without the need for further intervention. By tuning the dynamics of the processes, the molecular machine is able to move macrocycles from their lowest energy state in the solution to a higher energy state on the axle, maintaining out-of-equilibrium [n]rotaxanes as long as unreacted fuel is present.
