Recent loss of the Dim2 DNA methyltransferase decreases mutation rate in repeats and changes evolutionary trajectory in a fungal pathogen

Summary: In an important wheat pathogen, extensive differences in 5mC levels were found within a single species due to inactivation of the DNA methyltransferase Dim2, impacting genome stability and evolution. Presence of functional Dim2 was associated with high levels of 5mC in transposable elements, suggesting a role in genome defense, while strains with non-functional Dim2 alleles showed lower levels of 5mC maintained by Dnmt5. Functional Dim2 altered nucleotide composition by promoting C to T transitions at CpA sites during mitosis, contributing to transposon inactivation and impacting genome evolution in Z. tritici populations.

Fungal conidia prepare for the future by synthesizing and storing transcripts according to environmental conditions present before dormancy.
