To overcome these limitations, researchers have developed a bone marrow hydrogel that incorporates the complex and specific protein signature of bone marrow. This tissue-centric approach allows for better replication of the elasticity and structure of bone marrow, and provides a wide range of bone marrow-specific and cell-instructive peptides.

Research has shown that collecting and processing hematopoietic stem cells in a low oxygen (physioxia) environment can increase the collection of HSCs from bone marrow and other sources, potentially benefiting gene therapy and hematopoietic cell transplantation. Increased LT-HSCs in bone marrow from mice with Fanconi anemia collected and processed in physioxia may have clinical implications.

Summary: Recent research has shown that extracellular vesicles (EVs) secreted by adult bone marrow-derived mesenchymal stromal cells have the ability to support the ex vivo expansion of hematopoietic stem and progenitor cells (HSPCs), while those from fetal sources have a weaker effect. Protein composition analysis of EVs revealed that adult-derived EVs are rich in proteins related to oxidation-reduction processes, mitochondrial ATP synthesis, and protein folding, while fetal-derived EVs are rich in proteins linked to extracellular matrix organization, cell migration, and TGFBR signaling pathway.

This study uses microfluidic devices to create 3D hydrogels with lateral gradients in vascular density, revealing spatial patterns of HSC phenotype in response to angiocrine signals and showcasing the influence of local angiocrine signals on HSC fate.
