Spatiotemporal control of myofibroblast activation in acoustically-responsive scaffolds via ultrasound-induced matrix stiffening

Easton Farrell et al.

Summary: A novel composite hydrogel, called an acoustically-responsive scaffold (ARS), has been developed for non-invasive, spatiotemporally controlled modulation of mechanical and morphological properties using focused ultrasound. The study demonstrates that ARS can effectively control the differentiation of fibroblasts into myofibroblasts and induce matrix compaction and stiffening.

Stiffness increases with myofibroblast content and collagen density in mesenchymal high grade serous ovarian cancer

Virginie Mieulet et al.

Summary: Women diagnosed with high-grade serous ovarian cancers, especially with the Mesenchymal molecular subtype, show poor prognosis. These tumors exhibit distinct stiffness depending on their molecular subtype, with Mesenchymal tumors demonstrating a strong correlation between stiffness and tumor growth. Tumor stiffness is associated with stromal content, collagen network remodeling, and metabolic switch, potentially explaining the aggressiveness of Mesenchymal HGSOC.

A liquid culture cancer spheroid model reveals low PI3K/Akt pathway activity and low adhesiveness to the extracellular matrix

Natsuki Abe-Fukasawa et al.

Summary: This study examined liquid culture cancer spheroids for their apical-basal polarity, sensitivity to an Akt inhibitor, and interaction with ECM. The findings showed that these spheroids exhibit unique growth patterns and cellular characteristics under liquid culture conditions, which may have implications for cancer research and drug sensitivity testing.

Heterogeneous microenvironmental stiffness regulates pro-metastatic functions of breast cancer cells

Chun Liu et al.

Summary: The study showed that breast cancer cells behave differently in environments with different stiffness: cells tend to proliferate in soft environments and migrate in stiff environments. Breast cancer cells switch metabolic pathways and gene expression in response to varying environmental stiffness.
