Murine model of colonization with fungal pathogen Candida auris to explore skin tropism, host risk factors and therapeutic strategies

Xin Huang et al.

Summary: The study found that Candida auris establishes long-term residence within the skin tissue compartment, evading clinical surveillance, and clades of C. auris from different geographical origins differ in their abilities to colonize murine skin. IL-17 receptor signaling and specific arms of immunity protect mice from long-term C. auris skin colonization, and chlorhexidine antiseptic serves as a protective and decolonizing agent against C. auris.

Mrr1 regulation of methylglyoxal catabolism and methylglyoxal-induced fluconazole resistance in Candida lusitaniae

Amy R. Biermann et al.

Summary: Transcription factor Mrr1 regulates various genes including putative methylglyoxal reductases, which are important in metabolizing toxic metabolite methylglyoxal. Studies in Clavispora lusitaniae showed that Mrr1 regulates expression of MG reductases and also plays a role in fluconazole resistance. Endogenous and host-derived MG can induce MDR1 and other Mrr1-regulated genes causing increased drug resistance in Candida species, potentially contributing to fungal treatment failure.

Significant Differences in Host-Pathogen Interactions Between Murine and Human Whole Blood

Silke Machata et al.

Summary: By adapting the human whole-blood model to murine blood, it was found that murine blood has a weaker ability to reduce fungal burden and control filamentation of C. albicans compared to human blood. The lower neutrophil count in murine blood partially explains the insufficient infection control, and differences in host-pathogen interactions were observed, particularly in platelet interactions with C. albicans.

Summary: Candida auris has four major clades with different infection characteristics and drug resistance profiles. While Clade II primarily causes ear infections and is susceptible to antifungal drugs, Clades I, III, and IV are responsible for ongoing outbreaks of invasive and multidrug-resistant infections.

Environmental Isolation of Candida auris from the Coastal Wetlands of Andaman Islands, India

Parth Arora et al.

Summary: Studies have shown that Candida auris may be present in the marine ecosystem, and was isolated from salt marshes and sandy beaches near the Andaman Islands in the Indian Ocean. This suggests that prior to being recognized as a human pathogen, C. auris may have existed in the environment.

Balancing Positive and Negative Selection: In Vivo Evolution of Candida lusitaniae MRR1

Elora G. Demers et al.

Summary: The evolution of pathogens under selective pressures in chronic infections can impact their persistence, virulence, and response to antimicrobial therapy. Genetic analysis of isolates from single infections reveals dynamic selection pathways and trade-offs between high and low Mrr1 activity, affecting drug resistance and survival traits. Such dynamic evolution within chronic infections highlights the need for a balanced approach in managing microbial populations.

Candida pathogens induce protective mitochondria-associated type I interferon signalling and a damage-driven response in vaginal epithelial cells

Marina Pekmezovic et al.

Summary: Vaginal candidiasis is caused by four phylogenetically diverse species, each exhibiting distinct pathogenicity patterns during infection of vaginal epithelial cells. Host cells respond uniformly to all species in the early stages of infection, but diverge in a species-dependent manner later on, driven primarily by species-specific mechanisms of epithelial damage.

Candida auris Cell Wall Mannosylation Contributes to Neutrophil Evasion through Pathways Divergent from Candida albicans and Candida glabrata

Mark Horton et al.

Summary: Candida auris, an emerging fungal pathogen with high mortality rates, evades neutrophil activity through cell wall mannosylation, a mechanism distinct from other common Candida species. Disruption of mannosylation pathways renders C. auris susceptible to neutrophil killing and sheds light on innate immune evasion for this emerging pathogen.
