Exosomes from TNF-α-treated human gingiva-derived MSCs enhance M2 macrophage polarization and inhibit periodontal bone loss

Yuki Nakao et al.

Summary: Preconditioning gingival tissue-derived MSCs (GMSCs) with tumor necrosis factor-alpha (TNF-alpha) enhances the secretion of exosomes and the expression of CD73, promoting antiinflammatory M2 macrophage polarization. GMSC-derived exosomes exhibit significant ability to regulate inflammation and osteoclastogenesis, providing a potential therapeutic approach for periodontitis. The findings suggest a promising strategy for treating inflammatory osteoimmune disorders using TNF-alpha-preconditioned GMSC-derived exosomes.

Human mesenchymal stem cells promote ischemic repairment and angiogenesis of diabetic foot through exosome miRNA-21-5p

Chen Huang et al.

Summary: In this study, it was found that inflammatory cytokines and vascular cell adhesion molecule-1 induce MSCs to secrete exosomes heterogeneously. By RNA sequencing, a new proangiogenic miRNA, miRNA-21-5p, was defined. Knockdown and overexpression experiments validated the biological activity of exosome miRNA-21-5p, promoting in vitro cell proliferation, in vivo pro-angiogenesis in CAM assay, and tissue injury and repair angiogenesis in diabetic rat models. Exosome miRNA-21-5p was shown to promote angiogenesis through upregulation of VEGFR, and activation of AKT and MAPK pathways. This work suggests miRNA-21-5p as a potential novel mechanism for exosomes to promote ischemic tissue repair and angiogenesis in diabetic foot, with potential implications for clinical therapy.

Summary: MSCs, with their various chararcteristics and ability to differentiate into multiple cell types, possess a secretome that can modulate inflammatory responses and promote tissue repair/regeneration. Their immunomodulatory effects are achieved through the harmonious interaction of released signaling molecules and immune/target cells, making them potential therapeutic candidates for immune-mediated disorders.
